Objective:Breast cancer is the most common cause of cancer death in women at present.The incidence and mortality of breast cancer in our country increased every year,and the age of onset is becoming more and more younger.Research on breast cancer is also heating up,whereas the role of UPS in breast cancer attracted more and more attention of researchers;Studies have shown that,in the high glucose,the activity of UPS increased,and thus can produce the effect of promoting the development of MCF 7-cells.Deubiquitinating enzymes(DUB),an important part of UPS,by regulating the ubiquitin proteasome system,playing an important effect on the functional activity of cells.In the DUB has been found,ATXN3L has a strongly relationship with breast cancer;we speculate that in the high glucose,the promoted activity of MCF-7 cells is related to deubiquitinase ATXN3L.Auranofin,as a kind of deubiquitinase,could regulate the UPS and thus influence the effect of high glucose on the MCF-7 cells.This article will explore the effects of Aur acting on breast cancer MCF-7 cells in the high glucose and the potential mechanism.Methods:(1)The expression of ATXN3L in breast cancer cells MCF-7 at different concentrations of glucose:western blot was used to detect the expression of ATXN3L varying glucose concentrations.(2)The effect of Aur plays on the expression of ATXN3L in breast cancer cells MCF-7 at different concentrations of glucose:western blot was used to detect the expression of ATXN3L varying glucose concentrations after the treatment of Aur.(3)The effect of Aur plays on the proliferation,migration,invasion and apoptosis of MCF-7 cells at high glucose concentration:after Aur treatment,proliferation by MTT assay,migration by Scratch assay,invasion by Transwell assay and apoptosis of MCF-7 breast cancer cells by TUNEL assay.Results:(1)High glucose increases the expression of ATXN3L in breast cancer cells MCF-7.(2)The expression of ATXN3L is inhibited by Aur,and which is more apparent under the condition of high glucose.(3)High glucose-induc,ed promoting the proliferation,migration,invasion of MCF-7 cell were attenuated by Aur,as well as the inhibition of apoptosis.Conclusion:Our conclusion confirmed that Aur inhibits high glucose-induced promoting effect to MCF-7 cells,and this effect is probably by inhibiting the deubiquitinase to realize.
