| Objective: Breast cancer is the most common malignancy in women worldwide currently,and studies have shown that diabetes is a risk factor for breast cancer.More and more studies have found that BRCA1-associated protein-1(BAP1)is closely related to the development of various tumors,by regulating various processes such as protein ubiquitination degradation pathway,cell survival and apoptosis,DNA repair and cell cycle progression,but its effects on breast cancer cells and its specific mechanism remain to be explored.This study was to investigate the effects of BAP1 on the proliferation,migration and metastasis of breast cancer cell line MDA-MB-231 at different sugar concentrations and the possible mechanism so that maybe can provide new ideas for molecular targeted therapy of breast cancer.Methods:(1)Detection the expression of BAP1 protein in breast cancer cell MDA-MB-231 under different high glucose conditions.(2)To study the effects of deubiquitinase inhibitor Aur on the proliferation,migration,invasion,apoptosis and cycle of MDA-MB-231 cells at different sugar concentrations,and the expression of BAP1,ERK1/2,p-ERK,AKT/p-AKT and ER stress associated proteins.(3)To study the effects of BAP1 si RNA on proliferation,migration,invasion and apoptosis of MDA-MB-231 cells at different sugar concentrations.(4)The proliferation ability of MDA-MB-231 cells was detected by MTT assay.The migration ability was detected by Wound assay.The invasion ability was detected by Transwell assay,and the apoptosis and periodic changes were detected by flow cytometry.In the case,Western blot was used to detect the expression of various proteins.Results:(1)High glucose increased the expression of BAP1 in breast cancer cell MDA-MB-231.(2)Deubiquitinase inhibitor Aur can reverse the promotion of proliferation,migration and invasion of and MDA-MB-231 cells and inhibition of apoptosis to some extent.(3)Deubiquitinase inhibitor Aur can reduce the expression levels of BAP1 and ER stress associated proteins such as PERK,CHOP in breast cancer cell MDA-MB-231,and is more obvious under high glucose conditions.(4)BAP1 si RNA can reverse the promotion of proliferation,migration,invasion and apoptosis of MDA-MB-231 cells to a certain extent,and reduce the expression of BAP1 and ER stress associated proteins such as PERK,CHOP,and more pronounced under high sugar conditions.Conclusion: The deubiquitinating enzyme BAP1 can promote the proliferation,migration and invasion of breast cancer cell MDA-MB-231 and inhibit its apoptosis by regulating ER stress,and it is more obvious under high glucose conditions. |
