Objective: Primary liver cancer is a common global malignant digestive system of tumors with high morbidity and mortality,and is the third leading cause of cancer-related death.Due to the occult onset and faster progress,most patients have been in advanced and loss the chance of surgical resection?ablation or liver transplantation and other radical treatment.Transcatheter arterial chemoembolization(TACE)to be the main Palliative treatment.There is no uniform standard of circulating monitor for TACE in patients with liver cancer.At present,AFP is used as the index of biological detection.but its specificity is poor.Studies have shown that abnormal expression of micro RNAs is associated with various human-related cancers: including liver cancer,and miRNA expression profiles have demonstrated that the expression of miRNA-214 in hepatocellular carcinoma is down-regulated.So we detect the relative expression of miRNA-214 in serum of patients with primary hepatocellular carcinoma before and after TACE,and to explore the expression of serum miRNA-214 in serum of patients with hepatocellular carcinoma and the significance of short-term therapeutic effect after TACE.Methods: 30 HCC patients(experiment group),27 chronic liver disease(chronic hepatitis or cirrhosis)patients and 26 healthy volunteers were selected between January2016 and January 2017 in the second affiliated hospital of dalian medical university.The inclusive criteria of experiment group were composed of Untreated patient? No operative indications ? did not accept any other treatment and without the treatment taboo.The relative expression level of miRNA-214 in serum of the 30 HCC patients(before and after the TACE treatment)?27 chronic liver disease patients and 26 healthy volunteers was measured by quantitative reverse transcription polymerase chain reaction(q RT-PCR).At the same time,the patients in the liver cancer group were examined by imageological examination in 1 week before and 1 month after operation.The curative effect of the tumor was evaluated according to the improved curative effect evaluation standard(m RECIST).SPSS17.0 software was used to analyze the results.The experimental data were expressed as mean ± standard deviation.The difference expression of serum miRNA-214 in each group was compared,and the relationship between the differential expression of miRNA-214 and its clinical pathological parameters in liver cancer was also evaluated.Results: 1?The relative expression of serum micro RNA-214 in HCC patients before TACE(0.18±0.13)was significant compared to chronic liver disease patients and healthy controls(all P<0.05),while the differences of relative expression level of the two miRNAs between chronic liver disease patients and healthy controls was not significantly(P>0.05).2 ? The relative expression of serum microRNA-214 in HCC patients before TACE(0.18±0.13)was significant compared to the relative expression after TACE(0.57±0.09)(P<0.05)?3 ? The relative expression of serum micro RNA-214 in HCC patients after TACE showing partial response,stable disease and progressive disease were(1.28±0.17)?(0.95±0.30)and(0.18±0.01).The expression of serum micro RNA-214 in progressive disease(0.18±0.01)was significant compared to partial response,stable disease(all P<0.05),while the differences of relative expression level of the two miRNAs between partial response,stable disease was not significantly(P>0.05).4?The serum miRNA-214 level was closely correlated with the tumor size.Conclusion: The serum miRNA-214 show a significantly lower expression in patients with primary liver cancer.At the same time it also can well evaluating the effect of TACE.The serum miRNA-214 level in HCC patients was correlated with the tumor size.
