The Expression And Significance Of VEGF-A And LMP-1 In Peripheral T Cell Lymphoma

Background and Purpose: Peripheral T-cell lymphoma(PTCL)is a unique type of non-Hodgkin lymphoma(NHL),PTCL has many types of pathology,contains more than 20 unique types of diseases,it is characterized by distinct biological and clinical pathological features.Its pathogenesis and prognosis is related to many factors,EBV infection and tumor angiogenesis may have certain correlation with its pathogenesis and prognosis,in which,LMP-1 ? VEGF-A may play an important role in this process.Therefore,the aim of this study was to detect the expression of above proteins in PTCL,analyze the relationship between clinical prognosis indicators,and to explore the significance and correlation of VEGF-A and LMP-1 in PTCL.Methods: Select the formalin fixed,paraffin embedded specimens 31 cases,from PTCL patients between January 2008 and October 2016 in Second Affiliated Hospital of Dalian Medical University.VEGF-A and LMP-1 protein expression were detected in tumor tissue by streptavidin-peroxidase immunohistochemistry,and analyze the test results' correlation respectively with patients' age?gender?Ann Arbor features?B symptoms?LDH levels?IPI score?ECOG score.Statistical analysis was performed using SPSS statistical software package(version 22.0).The correlation of LMP-1 ?VEGF-A expression with contingency correlation analysis method,the VEGF-A andLMP-1 indicators associated with clinical prognosis correlation analysis by Fisher's exact probability method,P values lower than 0.05 considered as statistically significant.Result:1.The analysis of clinical features:31 cases of PTCL patients,all patients were initially treated,among them,9 NKTCL patients,22 PTCL-NOS patients;24 cases(77.4%)males and 7 females(22.6%),male to female ratio is 3.24:1;Age <60years old 17 cases(54.8%),?60 years old 14 cases(45.2%),with the median age is 58 years old(range 19-89 years);Most patients diagnosed with good physical condition,ECOG score:<2 points in 22 cases(71%),?2 points in 9 cases(29%);the serum LDH level is increased in 18 cases(58.1%),normal in 13 cases(41.9%);B symptoms in 14cases(45.2%),no B symptoms in 17 cases(54.8%);Ann Arbor stage: 12 cases of stage?-?(38.7%),19 cases of stage ?-?(61.3%);8 cases with stage I and II(26.7%),22 cases with III-IV(73.3%);the IPI score 0-2 points in 21 cases(low risk group 40%),3-5 points in 10 cases(high-risk group 32.3%),among the paitents younger than 60 years old were used aaIPI Grading standard.In 31 cases of PTCL patients,the immune group of VEGF-A is positive in 23 cases(74.2%),(+)7 cases,(++)8 cases,(+++)8 cases,8 cases of VEGF-A negative(25.8%);in patients with PTCL-NOS,VEGF-A positive expression in 19 cases,negative expression in 3 cases;in patients with NKTCL,VEGF-A positive expression in 8 cases,negative expression in 1 case;High expression rate was 51.6%(16/31),low expression rate was 48.4%(15/31),in patients with PTCL-NOS,VEGF-A high expression in 12 cases,low expression in 10,in patients with NKTCL,high expression in 4 cases,low expression in 5 cases.In 31 cases of PTCL patients,LMP-1 is positive in 12 cases(38.7%),18 cases of LMP-1 negative(61.3%);in patients with PTCL-NOS,LMP-1 positive expression in 9 cases,negative expression in 13 cases;in patients with NKTCL,LMP-1 positive expression in 3 cases,negative expression in 6 case.2.Statistical analysis showed:There have no statistically significant differences between the gender ? onset age ? Ann Arbor stage ? blood lactate dehydrogenase level?with or without B symptoms?ECOG score?IPI score with theexpression of VEGF-A?LMP-1(P>0.05).There have no correlation between VEGF-A expression and LMP-1 protein expression.Conclusion: 1.VEGF-A has higher positive expression rate in patients with PTCL,up to 61.3%.2.The positive expression rate of LMP-1 in patients with PTCL was 38.7%,which was similar to the reference.3.There have no statistically significant differences between the gender?onset age?Ann Arbor stage?blood lactate dehydrogenase level?with or without B symptoms?ECOG score?IPI score with the expression of VEGF-A?LMP-1(P>0.05).4.There have no correlation between VEGF-A expression and LMP-1 expression.
