Objective: Our country has a high prevalence of gastric cancer,whose incidence and mortality remain on a high level.Currently the major treatment is still surgery.The most cases are gastric adenocarcinoma,and for this pathological type,radiotherapy is not satisfactory.Therefore chemotherapy is the main auxiliary treatment.Even though early gastric cancer may acquire better prognosis with radical treatments,many patients were found in the progressive stage when they were admitted,due to the current hardship of early diagnosis and surveillance of progress,and a poor prognosis is indicated.Moreover,Chemotherapy may not offer satisfactory results,and it's toxic and side effects are significant.Therefore it is necessary to find new breaking points for early diagnosis,progress surveillance,and optimization of chemotherapy of gastric cancer.This research tested the expression level of NUMB and SIP1 in gastric tumor tissues and paracarcinoma tissues,and analyzed their mutual correlation and their correlation with clinical and pathological parameters,and analyzed their possible role in generation and development of gastric cancer,thus it may provide a new basis and starting point for early diagnosis and progress surveillance of gastric cancer,new targets for targeted medication and optimization of chemotherapy.Methods: 50 cases of gastric cancer samples,each with its paired paracarcinoma comparison obtained from surgical excision in Department of General Surgery of our hospital were selected.The expression of NUMB and SIP1 in gastric and paracarcinoma tissues were tested with immunohistochemical staining,and analyses for the correlation between the results and clinical / pathological factors,the correlation between NUMB and SIP1 expressions,and their roles in generation and development of gastric cancer were made.Statistical analysis was realized with SPSS 17.0 software,applying ?2 Test,Fisher Precise Test and Spearman Correlation Analysis.Statistical significance of the results was marked by P<0.05.Results:1.The Contrast of expressions of NUMB protein in gastric tumor and paracarcinoma tissuesIn these 50 cases of gastric cancer,the high expression rate of NUMB in tumor tissue is 8.00%,while in paracarcinoma tissues;the high expression rate is 88.0%.The expression of NUMB in paracarcinoma tissues is higher than in tumor tissues obviously.The differentiation has a statistical significance(?2=60.938,P<0.05).2.The Contrast of expressions of SIP1 protein in gastric tumor and paracarcinoma tissuesIn these 50 cases of gastric cancer,the high expression rate of SIP1 in tumor tissue is 86.00%,while in paracarcinoma tissues;the high expression rate is 36.00%.The expression of SIP1 in tumor tissues is higher than in paracarcinoma tissues obviously.The differentiation has a statistical significance(?2=24.212,P<0.05).3.The correlations between NUMB expression and various clinical / pathoilogical parametersThe expression of NUMB shows no statistical significance,between the male and the female,between ages >55 years old and ages ?55 years old,between Stage I-III and III-IV in terms of TNM stages,between T1-2 and T3-4 in terms of infiltration,between diameters <5cm and ?5cm,between with and without lymph node metastasi,between low differentiation and medium to high differentiation,between intestinal and diffuse types in Lauren typing(P>0.05).4.The correlations between SIP1 expression and various clinical / pathological parametersThe expression of SIP1 shows statistical significance between,between groups of T1-2 and T3-4 in terms of infiltration(?2=3.476,P=0.039<0.05),while between the male and the female,between ages >55 years old and ages?55 years old,between Stage I-III and III-IV in terms of TNM stages,between diameters <5cm and ?5cm,between with and without lymph node metastasis,between low differentiation and medium to high differentiation,between intestinal and diffuse types in Lauren typing,the expression of SIP1 shows no statistical significance(P>0.05).5.The correlation between expression levels of NUMB and SIP1 in gastric cancer tissuesThe expression level of NUMB protein compared with that of SIP1 shows a negatively correlated relation(r=-0.306,P=0.030<0.05).Conclusion:1.The expression of NUMB in gastric cancer tissues is weakened,while the expression of SIP1 is elevated.The two expression levels show a negative correlation.2.The expression level of NUMB shows no correlation with gender,age,TNM stages,tumor infiltration,size of tumor,differentiation,or Lauren typing.3.The expression level of SIP1 shows a significant correlation with tumor infiltration,while there is no correlation with gender,age,TNM stages,size of tumor,differentiation,or Lauren typing.4.There exists mutual effects between NUMB and SIP1,thus making them capable to serve as teamed-up markers for fast diagnosis of gastric cancer,in order to promote the rate of early detection.SIP1 can be used for development surveillance of gastric cancer,and can serve as a new target for targeting anti-cancer drugs,providing a new start line for inhibition of tumor invasion,metastasis,and recurrence,and for promoting the effects of chemotherapeutic drugs.
