Objective:To observe the expression levels of IFN-?1 in children with RSV infection of different viral load,analyze the relationship between the expression levels of IFN-?1 and the disease course,reveal the role of IFN-?1 in RSV infection.Method:1.The children nasopharyngeal swab were collected from the sick children in Hunan Provincial People's Hospital pediatric outpatient and inpatient and the healthy children for physical examination in June 2015-June 2016,using direct immunofluorescence method to detect the seven common respiratory virus antigen(include RSV).Select the only RSV positive samples for infectious group(120 cases),non-positive samples for healthy controls(50 cases).The RSV mRNA,IFN-?1mRNA and GAPDH mRNA in samples were detected using Quantitative Real-time PCR method.Statistical analysis of the relationship between the expression levels of each mRNA.2.useing the electronic medical records system to obtain the basic information and the condition of RSV infection in group,statistical analysis the expression level of IFN-?1 and the severity of the disease,understand the effect of IFN-?1 on the condition of RSV infection.Results:1.In RSV infected children,79%of the children were only RSV positive,and 21%of children with other pathogens infection.Mycoplasma pneumoniae was the most common pathogen in the combined infection.2.In RSV infected children,The disease severity was associated with the viral load.(t=6.41,P<0.05)3.The expression level of IFN-?1 in RSV infection group was significantly higher than that in healthy control group,the difference was statistically significant(t=6.64,P<0.05).And there has no relation with age and sex.4.There was a positive correlation between the expression levels of IFN-?1 and RSV loads(r=0.56,P<0.05).5.The expression level of IFN-?1 was not significantly correlated with the duration of wheezing in children with RSV infection(r=-0.285,P=0.238).6.In the same level of the RSV loads,there was a negative correlation between the IFN-?1 expression and hospitalization time.(the viral load was divided into ×104?×105?×106?×107,and the correlation coefficients were-0.743,-0.470,-0.601,-0.635,respectively;the P values were respectively:0.004,0.012,0.001,and 0.008,respectively.)Conclusion:l.The severity of RSV infection in children was related to RSV load.2.Respiratory syncytial virus can induce the expression of IFN-X1 in respiratory epithelial cells of children,and the body can also indirectly influence the process of RSV infection by regulating the expression of IFN-lambda 1.
