ObjectivemicroRNAs(miRNAs)are a class of small non-coding RNAs,which post-transcriptionally regulate gene expression.miRNAs are involved in many physiological and pathological progress.More and more evidences indicate that miRNAs may function as oncogenes or tumor suppressors and play an important role in tumorigenesis by regulating their target genes.In our previous study,we found that traditional Chinese medicine compound Yangzheng Sanjie Decoction(YZSJD)can significantly inhibit gastric cancer cell proliferation and promote apoptosis of gastric cancer cells.On the basis of the previous research,we explore the influence of YZSJD on miRNAs,and the possible molecular mechanisms of YZSJD mediated by tumor suppressor-miRNAs to regulate the growth of gastric cancer cells.Methods1.using miRNA chip inspection to detect the difference expression of miRNAs after YCS and the control group treatment in SGC-7901 cells.2.representative tumor-suppressor miRNAs let-7a and miR-7 were chosen for further verification in AGS,SGC-7901,HS-746T and MKN-45 cells by using the q RT-PCR method.3.using miR-7 mimic to overexpress miR-7 and miR-7 inhibitor to inhibit the expression of miR-7,CCK-8 assay to detect the effects of miR-7 on gastric cancer cell proliferation,wound healing and transwell assay testing the effects on gastric cancer cell migration.4.detect c-Myc/Ras protein expression which is down-regulated by let-7a,EGFR protein expression which is down-regulated by miR-7 in the YCS group and the control by western blot.Results1.The miRNAs expression is differently expressed after YCS treatment,compared with the control group.2.After YCS treatment,let-7a and miR-7 expression are significantly up-regulated in AGS,SGC-7901,HS-746T and MKN-45 cells,compared with the control group(P<0.05).3.Inhibition of gastric cancer cell proliferation and migration are induced by miR-7(P<0.05).4.By comparison with the control group,the expression of Ras,c-Myc and EGFR are significantly lower in YCS group in GC cells(P<0.05).ConclusionYCS changes the miRNAs expression in gastric cancer cell,and miR-7/EGFR,let-7a/c-Myc/Ras signal pathway may be the possible mechanisms of YZSJD regulating the gastric cancer cell growth mediated by tumor-suppressor miRNAs.
