Objective1.Using immunohistochemistry technique to detect the expression of Vasohibin-1 and Haparanase in the different stages of intantile dermal capillary hemangioma.2.To investigate the mechanism and significance of Vasohibin-1 and Haparanase in the occurrence,development and regression of infantile dermal capillary hemangioma and analyze the relationship between Vasohibin-1 and Haparanase in oder to know the impact of the prognosis of infantile dermal capillary hemangioma,which can provide a new direction and new ideas in the clinical prevention,diagnosis and treatment of infantile dermal capillary hemngioma.Methods1.To collect paraffin specimens of hemangiomas and clinical hospitalizing data from January 2014 to January 2016 who just accepted operations without any other therapies at the department of Aesthetic Plastic Surgery of Ren Ming Hospital of Wuhan University,52 in total.All specimens have been diagnosed by the pathologists in the hospital,selecting 5 cases of normal skin tissue adjacent to hemangiomas as control group.According to Mulliken's classification and the expression of proliferating cell nuclear antigen(PCNA),52 specimens were classified to 28 proliferating phase capillary hemangiomas and 24 involuting phase capillary hemangiomas.Using immunohistochemistry technique and dyeing method to test the expression level of Vasohibin-1 and Haparanase in each phase and the group of normal skin tissue.2.Analyze each photo with the aid of computer Image-Pro Plus 6.0 software.Determine and Calculate the average optical density value and the positive rate of area of Vasohibin-1 and Haparanase by semi-quantitative analysis.Use SPSS 16.0 statistical software to deal with the measured data and analyze the differences among all groups and detecte the correlation between Vasohibin-1 and Haparanase.The difference was statistically significant(P<0.05).Result1.The expression of VASH-1Positive expression of VASH-lwas located in the cytoplasm of hemangioma endothelial cell,brownish or brownish yellow;The weakly positive or negative expression in proliferating phase and normal dermal tissues,strongly positive expression in involuting phase.The average optical density in two phases and the group of normal skins were 0.0042±0.0012?0.0157±0.0011?0.0016±0.0015 respectively.The mean positive rate of area in two phases and the group of normal skins were 0.3493±0.0241?4.5877±0.0242?0.0447±0.0284 respectively.The expression of VASH-lin involuting phase is significantly higher than that in proliferating phase.The differences were statistically significant(P<0.05).The expression in proliferating phase is higher than that in normal skin group.The differences were statistically significant(P<0.05).2.The expression of HPAPositive expression of HPA was located in the cytoplasm of hemangioma endothelial cell,brownish or brownish yellow;The weakly positive or negative expression in involuting phase and normal dermal tissues,strongly positive expression in proliferating phase.The average optical density in two phases and the group of normal skins were 0.0146±0.0003?0.0032±0.0002?0.0022±0.0003 respectively.The mean positive rate of area in two phase and the group of normal skins were 2.2780±0.0096?0.5226±0.0157?0.0250±0.0150 respectively.The expression of HPA in proliferating phase is significantly higher than that in involuting phase.The differences were statistically significant(P<0.05).The expression in involuting phase is higher than that in normal skin group.The differences were statistically significant(P<0.05).3.Correlation analysisThe average optical density:There is a significant negative correlation relationship between the expression of VASH-1 and HPA(r =-0.723 P<0.01).The average positive rate of area:There is a significant negative correlation relationship between the expression of VASH-1 and HPA(r =-0.724 P<0.01).ConclusionThe expression of VASH-1 in proliferating phase is obviously higher than in involuting phase,which indicates this factor may be involved in regulation of infantile dermal capillary hemangioma endothelial cell apoptosis process.The expression of HPA in involuting phase is obviously higher than in proliferating phase,which indicates this factor may play an important role in the infantile dermal capillary hemangioma endothelial cell proliferating process.Both of them influence each other,work together to promote the occurrence and development of hemangioma and degradation.
