Objective: To prove the suppressive effect of anti-KDR antibody against vascular endothelial growth factor (VEGF) on proliferation of vascular endothelial cells of hemangioma via combining VEGF receptor 2(KDR) with anti-KDR antibody in competition with VEGF.Method: Five hemangiomas of proliferative phase from 5 children was cultured (HVEC) via using tissue culture method. Both primary culture and sub-culture were conducted in M199. The HVECS of passage 3 were divided into 4 groups based on the various concentrations of anti-KDR antibody added to the M199 medium where the HVECS were cultured. Group 1, free interference, namely control group; group 2, interfered with lower concentration of anti-KDR antibody; group 3, interfered with medium concentration of anti-KDR antibody; group 4, interfered with higher concentration of anti-KDR antibody. Cell count (CC) and the inhibitor rate of HVECS proliferation were conducted respectively within 48 hours after interference.Results: The number of HVECS of the three interfered groups was sharply dropped. The inhibitory rate of anti-KDR(50ug/ml, 10 ug/ml, 2 ug/ml) was 84%, 63%,39%, which showed that inhibitory effects correlated positively with the concentration of anti-KDR. On the contrary, the number of the free-interference group (control group) increased significantly.Conclusion: The anti-KDR antibody may suppress the action of VEGF through blocking the VEGF receptor 2(KDR), which shows us the potential clinical application in the treatment of hemangioma where pathological angiogenesis is involved.
