Objective Observing the expression of PACER and NF-?Bp50 and COX-2 in the renal tissue of rats with uric acid nephropathy,and to detect the urinary protein,UA,Urea,Creatinine(Cr),observe the pathological changes of renal tissue in rats with uric acid nephropathy,including inflammation and fibrosis pathological changes,and the treatment of uric acid nephropathy with Chinese medicine Weicao capsule to observe the impact of uric acid nephropathy in the above indicators.To explore the specific mechanism of Weicao Capsule to reduce blood uric acid,reduce inflammatory reaction,delay renal fibrosis and protect renal function,and provide theoretical basis for clinical treatment of uric acid nephropathy with Weicao capsule.MethodsExperiment one:20 SPF SD male rats were randomly divided into normal control group and model group.The rats in model group were fed with normal diet,and the rats in model group were fed with urinary creatine powder and yeast extract.The rats were sacrificed 24 hours later and the urine protein was collected.The levels of UA,Urea and Scr were measured in the vena cava.The expression of PACER and NF-?Bp50 mRNA and COX-2 mRNA were detected by RT-PCR in HE staining and Masson staining.The expression of NF-KBp50 and COX-2 protein was detected by western blot,and compared with normal control group.Experiment one:60 SPF grade SD male rats were randomly divided into low dose group of Weicao Capsule,middle dose group of Weicao Capsule,high dose group of Weicao Capsule,etoricoxib group and model group,and normol control Group,each group of 10,the normal group to give the general feed feeding,the other five groups to give adenine + yeast powder special feed feeding,feeding 19 days later,each rat through the orbital canthus vein blood 0.5 ml,measured serum UA value,The blood uric acid in rats fed the diet was higher than 82.5?mol/L,indicating that the model of hyperuricemia was successful.At the same time,the urine was collected and the urine protein concentration was measured.The groups fed with special feed were significantly higher than those in the normal control group,suggesting that the kidney was damaged and the model of uric acid kidney damage was successful.The rats in the normal control group and the model group were treated with distilled water 10ml/kg daily.The low dose group,middle dose group and high dose group were given 0.3g,0.6g,1.2g/kg Weicao capsule,for 6 weeks of administration;etoricoxib group were given etoricoxib 10 mg/kg cough.Blood samples were collected from the 4th week and the 6th week respectively.24h urine protein was measured and 0.5ml blood was taken from the orbital inner canthus.The levels of UA,BUN and Scr were measured in each group.The rats were treated in the same way,and the corresponding indexes were measured and compared with the normal control group,the model group,etoricoxib group and low dose group of Weicao CapsuleResultsexperiment one:1.Compared with the normal control group,the levels of 24 hours urinary protein,serum UA,Urea,Scr in the model group were significantly higher than those in the control group(P<0.01).2.HE staining Compared with normal control group,renal tubular sections of the model group showed atrophy,luminal enlargement,renal tubular,renal interstitial inflammatory cell infiltration,changes in fibrous proliferation.Masson staining Compared with the normal control group,the model group increased the number of tubular fibrosis and the degree of fibrosis was heavy.3.Compared with the normal control group,the expression of IncRNA PACER,NF-?Bp50,COX-2 protein and its mRNA were significantly increased in the model group(P<0.05).Experiment two1.Biochemical indicatorsCompared with the normal control group,the levels of urinary protein and serum UA,Urea and Cr in the model group were significantly higher than those in the normol control group(P<0.05).After 4 weeks of treatment,compared with the model group,(P<0.05).Compared with the control group,the UA,Urea and Cr levels of the rats in the group were significantly higher than those in the control group(P<0.05)(P<0.05).The levels of UA,Urea and Cr in the medium and high dose groups were significantly better than those in the low dose group(P<0.05).After 6 weeks of treatment,compared with 4 weeks after treatment,the dose of 24 h urinary protein and the improvement of serum UA,Urea and Cr were better,the difference was statistically significant(P<0.05)The above indexes were higher than those in the fourth week,and there was statistical difference(P<0.05).2.HE staining,Masson stainingUnder the light microscope,HE staining showed that the renal tissue of the model group had atrophy,luminal enlargement,renal tubular,renal interstitial inflammatory cell infiltration,renal interstitial fibrosis-like changes;The number of inflammatory cells in the high dose group and the control group was lower than that in the model group,and the number of renal tubular atrophy was decreased and the renal interstitial fibrosis was less changed.The small dose group was similar to the model group.Masson staining in the model group:the type of renal tubular fibrosis in the model group increased,the degree of fibrosis was severe.After treatment,the dose group and the high dose group of Weicao Capsule were less fibrotic area There was no significant change in the low dose group compared with the model group.3.The expression of IncRNA PACER?NF-?Bp50?COX-2Compared with the normol control group,the expression of PACER,NF-?Bp50 and COX-2 protein and mRNA in the renal tissue of the other groups were significantly higher than those in the control group(P<0.05).The expression of PACER,NF-?Bp50 and COX-2 protein and mRNA in the Weicao capsule middle dose group,Weicao capsule high dose group and etoposide group were significantly lower than those in the control group(P<0.05).The expression of PACER and NF-?Bp50 and COX-2 protein and mRNA in the Weicao capsule medium and high dose groups were significantly lower than those in the control group(P<0.05)2 protein and mRNA levels were lower than those in the low dose group(P<0.05).conclusion1.The rats of urinary acid nephropathy have a certain degree of renal damage,Weicao capsule can reduce 24h urinary protein,serum UA,Urea,Cr levels,improve renal function,long-term take better;Time to improve renal function,taking a long time but damage kidney function.It can be seen,Weicao capsule has smaller side effects than etoposide,and more safety.2.The activation of IncRNA PACER,NF-?Bp50 and COX-2 in the kidneys of uric acid nephropathy rats was inhibited by Weicao capsule,presumably to prevent the expression of IncRNA PACER and inhibit the expression of NF-?Bp50 and COX-2 Is one of the important mechanisms of Weicao Capsule to inhibit renal inflammation and reduce renal interstitial fibrosis.
