Bifidobacterium was first isolated from the feces of breastfed infants by Henri Tissier in 1899.Bifidobacterium was continuously studied as an important probiotic for more than a century after being discovered.Nowdays,it has been widely used in the production of dairy and microecological preparations.As human and animal intestinal important probiotics,bifidobacteria is closely related to the body of many physiological and pathological phenomena and considered as one of the important indicators of human health.However,Bifidobacterium as a strict anaerobic bacteria,they have harsh culture conditions and slowly growth,in addition to difficult to store.Therefore,how to promote the proliferation of bifidobacteria in vivo or in vitro is one of the key issues for its application.The nutrient requirements of bifidobacteria is high,so it often adds exogenous growth factors in vitro culture.These substances which have a role in promoting bifidobacteria called bifidus factor.Nowdays,Oligosaccharides and polysaccharides etc.are the most commonly added.Traditional Chinese medicine is a medical treasure house,embodies intelligence of chinese people and has made great contribution to human health,which has a wide range of functions,two-way regulation,less side effects,no drug resistance and other advantages.Tonic traditional Chinese medicine is a class of drugs of which with tonic righting and correcting the body qi and yin deficiency pathological bias.Modern researches show that tonic traditional Chinese medicine have immune regulation,anti-aging and other physiological activities.In this study,we studied the effects of various traditional Chinese medicine on the growth of bifidobacteria in vitro and their synergistic anti-tumor effect,which in order to provide new ideas for further development and utilization of traditional Chinese medicine resources and bifidobacterium products.At first,we studied the effects of the twelve kinds of tonic traditional Chinese medicine on the growth of Bifidobacterium longum in Vitro,including Lilii bubus,Aractylodis macrocepHalae rhizome,Glehniae radix,Asparagi rhizome,Pseudostellariae radix,OpHiopogonis radix,Lycii fructus,Astragali radix,Codonopsis radix,Ecliptaeherba,Acanthopanacis senticosi radix etc.At the same time,the growth curves of Bifidobacterium longum effected by the different concentrations of those traditional Chinese medicine were recorded and compared.The results showed that the promoting effect of Pseudostellariae radix and Astragali radix on bifidobacteria were the most significant and stable.On the basis,re-preparation water extract of Pseudostellariae radix and Astragali radix was to further determine the effect of Pseudostellariae radix and Astragali radix on bifidobacteria.Then we specific researched the effect of Pseudostellariae radix and Astragali radix on three strains of bifidobacteria and different concentrations of liquid.Compared with the control group,the results showed that Pseudostellariae radix and Astragali radix have a significant promotion effect on Bifidobacterium longum,Bifidobacterium animalis subsp.lactis and Bifidobacterium infantis in a concentration-dependent relationship.The higher the concentration,the better the promotion effect.But by analyzing the growth curve,the differences can be found in the specific mode of action.Astragalus had no significant promotion effect on the growth rate of bifidobacteria,but it could significantly prolong the time of bifidobacteria reaching the plateau,thereby increased the bacterial counts.However,the promotion effect of Pseudostellariae is not only to increase the growth rate of bifidobacteria and extend the logarithmic growth period.After that,we innovative explore the combination of Astragalus and Pseudostellariae with bifidobacteria for the treatment of melanoma in mice,the results showed that Astragalus and Pseudostellariae combination wth bifidobacteria can significantly improve the tumor-bearing mice tumor inhibition,and the more significantly prolong the survival of tumor-bearing mice time compared with administration of the two separately.Then we preliminary study the mechanism of the effects of Pseudostellariae radix on bifidobacteria from the perspective of chemical composition.Using rapid high performance liquid chromatograpHy,the contents of 16 amino acids,12 nucleosides and 2 cyclic peptides in Pseudostellariae radix were determined.The results are shown that arginine is the highest content of Amino acid composition,followed by aspartic acid.The content of guanosine in nucleoside is the highest,followed by adenosine.Cyclic peptide B is higher than cyclic peptide A in cyclic peptide in the sample.The actual content of several components with the highest content in the aqueous extract of Pseudostellariae radix was calculated.The same content of the standard product was added to the three strains of bifidobacteria in the medium,comparison with Pseudostellariae solution and control.The results found that the promotion effect of the raw materials is significantly better than anyother compositions,which means that the promotion effect of Pseudostellariae is not caused by a single component.In general,amino acid components play a role in promoting the role,but its promotion effect is far less than the liquid,it showed that there are other ingredients work.
