Objective:To establish methods for the determination of 4-hydroxyisoleucine,saponin and Trigonelline in Trigonella foenum-graecum L.seeds and Trigonella foenum greacum.L extract of alcohol?TFGe?by TLC,UV and HPLC.To observe protective effects of TFGe on chemical liver injury in mice.Method:Using the CHP method to determinate trigonelline.4-hydroxyisoleucine adopted precolumn derivatization method of HPLC,using phenylisothiocyanate as derivative reagen.Using the UV method to determinate saponin.Taking different drying technology through the detection of4-hydroxyisoleucine and Trigonelline,optimum extraction process.The model of chemical liver injury was established by tail intravenous injection with concanavalin A?ConA?,and normal control group getting equal volume of normal saline,the positive drug control group use silibinin.Different dosages of TFGe and TFGe tablets were given respectively.At the end of the experiment,the model group and each sample groups received intravenous dose of 15mg·kg-11 ConA(10mL·kg-1 BW).The concentration of the serum Alanine aminotransferase?ALT?,Aspartate aminotransferase?AST?,Lactate dehydrogen?LDH?,Reactive Oxygen Species?ROS?were measured by extracting the eyeball blood after fasting 8h.Results:A good linearity was obtained in the range of4.220.9?g·mL-1?r=0.9993?for 4-hydroxyisoleucine.The average recovery of 4-hydroxyisoleucine was 96.8%,RSD=1.65%.The liner range of saponin was 0.050.25mg?r=0.9995?,the average recovery was 96.6%,RSD=1.16%.In vacuum low temperature drying conditions,The efficacy contents are stable.The maximum tolerance dose of TFGe is larger than 40g·kg-1 in mice.With TFGe treatment,The concentration of the serum ALT,AST,LDH,ROS were significantly depressed as compared with that of the model group.TFGe could significantly improve the liver cell degeneration in mice with chemical liver injury.Conclusion:Using phenylisothiocyanate as derivative reagent,the precolumn derivatization methods of HPLC is simple,reproducible and accurate.The HPLC method could be used in the quantitative determination of Trigonella foenum-graecum L.seeds and TFGe.TFGe extraction process is simple and efficient.The satfty taking TFGe is high.TFGe and TFGe tablets could reduce the enzyme,and have protective effects on chemical liver injury in mice.
