4-hydroxyisoleucine is a kind of non-protein amino acids in Fenugreek seeds, it can promote insulin secretion, regulating blood glucose and blood lipid, which is one of the main active ingredients that Fenugreek hypoglycemic lipid-lowering effect. Many reports on the pharmacological effects of 4-hydroxyisoleucine and reducing blood glucose mechanism, but lew reports in the preparation process. In order to better development the medicinal resource, this paper intends to find a preparation process for 4-hydroxyisoleucine extracts and monomer from Fenugreek seeds, which is suitable for industrial production. It can provide and technical process for the subsequent development and research on the drugs to treatment ol type 11 diabetes.The lirst part of the thesis studies the preparation process of 4-hydroxyisoleucine extract from Fenugreek extracts. First we established a method for determination of the content of 4-hydroxyisoleucine by HPLC, and using different kinds of materials lo do a research. The results showed that the content of 4-hydroxyisoleucine in Fenugreek extracts by ADS-5 macroporous resin can be increased significantly, and it can be used for preparation of 4-hydroxyisoleucine extracts, finally get the extracts content of 4-hydroxyisoleucine more than 35%, and the operation steps are simple and quick.The second part of the thesis focused on the study of preparation process of 4-hydroxyisoleucine monomer from Fenugreek extracts, the method by amino acids precipitation, method of BOC protected amino acids are not suitable, and method of lactonization of 4-hydroxyisoleucine successfully prepared of 4-hydroxyisoleucine monomer, it can be used for industrial production.At last, the information about the effective components in reducing blood glucose of fenugreek seeds and the ways of 4-hydroxyisoleucine lo improve insulin resistance glucose and lowering blood glucose mechanism were reviewed.
