ObjectiveSepsis is an infection caused by uncontrolled systemic inflammatory response syndrome,which even can lead to death.The inflammation is associated with pathogenes of many diseases such as aging?cancer and the iscardiovascular system disorders.Sanguinarine is one of the active ingredient in the taditional chinese medicine plant Macleaya cordata(Willd.)R.Br.,It belongs to a kind of isoquinoline alkaloids.According to the recent study,sanguinarine has funtion of anti-tumor?antimicrobial insecticidal.In order to explore the further pathogenesis on anti-inflammatory and the mchanism,we used LPS stimuliated the macrohages and mouse,studied the sanguinarine effcet on NF-?B and STAT3 signaling pathways,for the next experiments of sanguinarine.MethodsThis study will explore the influence of sanguinarine associated with systemic inflammatory response induced by LPS by using the cell experiments and the animal experiments.The cell experiments1 To find out the sitable concentration of sanguinarine,the MTT was used to the different concentration of sanguinarine on the RAW264.7cell or the LPS induced RAW264.7cell.2 After 24h of the sanguinarine with the LPS induce RAW264.7cell,the supernatant was selected,enzyme linked immunosorbent assay was used to detect the concentration of IL-6 and TNF-?.3 After 24h of the sanguinarine with the LPS induce RAW264.7cell,the cell was celected with trozil,then the RT-PCR used to text the expression of IL-6?TNF-??iNOS?STAT3?NF-?B p65 in the cell.4 Western blot was used to the protein expression of NF-K B signaling pathway and the STAT3 signaling pathway.The animal experiments1 the ALT/AST concent of serumin the mouse was texted by using kits,the concent of IL-6 and TNF-? texted by enzyme linked immunosorbent assay.2 the tissue pathological changes were observed Through hematoxylin-eosin assay.3 The mRNA of IL-6?TNF-??iNOS?STAT3?NF-K B p65 in the liver and lung was detected by RT-PCR.4 Western blot was used to the protein expression of NF-K B signaling pathway and the STAT3 signaling pathway in the liver and lung.ResultsIn the viro experiments,the results showed that the sanguinarine could reduce the concernation of IL-6 and TNF-?,the mRNA of IL-6?TNF-??iNOS?STAT3?NF-?B p65 and the protein expression of NF-?B p65?I?B??JAK2 and STAT3 was decreasedin the cell.In the vivo experiments,sanguinarine could improve pathological changes,the concenation of AST?ALT?IL-6?TNF-? was reduced by sanguinarine.Sanguinarine decreased the protein expression of NF-?B p65?I?B??JAK2 and STAT3 in the liver and lung.According to the experiments,the data showed that the sanguinarine has a positive effect to LPS induced inflammation with a dose-dependent manner.ConclusionAccording to the result,sanguinarine has obvious inhibitory on the protein expression of NF-?B signaling pathway and the STAT3 signaling pathway,and sanguinarine could increased the pathological changes,so the effect of sanguinarine on the inflammation induced by LPS has a corresponding relationship with NF-?B signaling pathway and the STAT3 signaling pathway.
