Sanguinarine is benzo[c]phenanthridine alkaloid with anti-inflammatory effect. Researches on the mechanism of sanguinarine anti-inflammation have very potential importance for exploring new anti-inflammatory drugs.Methods:Thirty two rats were randomly divided into 4 group(each has 8 rats):the control group, the model group,the positive drug group (SASP) and the sanguinarine group. According to the feeding model,the control group was fed normally. After 7 days of drinking distilled water, the model group, the positive drug group (SASP) and the sanguinarine group drinked 5% DSS water solution, meanwhile the positive drug group and the sanguinarine group were intragastrically administrated with nitrogen methyl pyridine (SASP) 225mg/kg.d and sanguinarine 75mg/kg.d from the first day to 14th day, respectively. From 8th day weight loss, stool consistency,blood or gross bleeding and disease activity index (DAI) of rates in each group were detected every day. At 15th day of experiment the rates were sacrificed and the colon and serum of each rate were sampled. Expression of proinflammatory cytokines (IL-1B, IL-6, IL-10 and TNF alpha) in colon were detected with qPCR. Expression of these proinflammatory cytokines in serum were also detected with ELISA. Colon was histopathologically detected by H.E and MyD88 Expression and NF-?B(p65) activity by immunohistochemistry.Results:DSS caused fine enteritis. The expression of IL-1B, IL-6 and TNF-a cytokines in colon of rats administrated with Sanguinarine and SASP decreased, but expression of IL-10 increased. These inflammatory factor levels in serum and colon revealed no obvious changes. Goblet cells and glandular cells of rats administrated with Sanguinarine and SASP increased. MyD88 Expression and NF-?B(p65) activity in colon of these rats increased compared with control groups.Conclusion:The results of this study suggested that the anti-inflammatory effect of sanguinarine was mediated through NF-kappa B signaling pathway.
