The Effects Of Lycium Barbarum And Salvia Miltiorrhiza On The Histomorphology And CRYAB MRNA Of Retinal Pigment Degeneration Rats

Objectives:To observe the effects of Lycium barbarum and Salvia miltiorrhiza on the histomorphology and CRYAB mRNA of retinal pigment degeneration rats.Methods:24 rats(48 eyes,male and female)with RCS(rdy-/-,p-/-)were randomly divided into two groups,each group of 8 rats(16 eyes),male and female half.It is the model group,the Chinese wolfberry and Salvia miltiorrhiza treatment group(Qishen group).The other 8 rats(16 eyes)with RCS(rdy+/+,p+/+),male and female half,as the blank group.We made 3groups:blank group,model group,Qishen group.We began the gavage after routine feeding for 1W,1 /d.The blank group and model group were given normal saline,Qishen group was given the equivalent dose of medlar and Salvia miltiorrhiza.We put the rats to death after continuous intragastric for 30 days.We observed the morphological changes of the thickness of the outer nuclear layer of all the rats with RCS under light microscope and detected the expression of CRYAB mRNA in the retina of RCS rats by qPCR,then analyzed the results.Results:(1)The retina HE staining results of the rats in each group showed: The retinal structure of each group in the blank group was arranged orderly and orderly.Compared with the blank group,the retinal structure of the model group was obviously loose,degeneration and atrophy.Compared with the blank group,the Qishen group was more disordered and the thickness was thinner,but the difference was statistically significant compared with the model group(P < 0.01).(2)The expression of CRYAB mRNA was found in the retina in the blank group,the expression of CRYAB mRNA in retina of rats with RP lesions was significantly lower than that in RCS group(P<0.01),the expression of CRYAB mRNA in retina of rats fed with medlar and Salvia miltiorrhiza was higher than that in model group(P<0.01),and it had a high statistical significance.Conclusions:(1)The RCS rat model was used as an animal model of retinal pigment degeneration,which can simulate the onset of retinitis pigmentosa very well;(2)Retinitis pigmentosa of RCS rats in various layers of retinal degeneration,the outer nuclear layer became thinner,and the expression of CRYAB mRNA was inhibited.(3)Lycium barbarum and Salvia miltiorrhiza can induce the expression of CRYAB mRNA in retina,and have protective effects on the retina of RCS rats.
