Bitter almonds are good medicinal and edible plants,with a variety of biological activities of anti-cancer,anti-tumor,embellish aperient bowel and antihyperglycemic.Amygdalin,one of the main component of bitter almond,has large market demands.Thus,on the basis of literature review and a large number of experiments,a method for defatting and extracting amygdalin by 95% ethanol is developed.The main research contents include:Amygdalin determination method: A HPLC determination method of amygdalin was optimized and validated,which provided the basis for the follow-up amygdalin determination.Extraction and purification technology: Conduct a comprehensive study on the enzyme,grinding,extraction,degreasing,crystallization,purification,drying and other steps of bitter almond.A method for defatting and extracting amygdalin by 95% ethanol directly is developed.The extraction technology was optimized by single factor experiment and response surface method.The optimized extraction rate of amygdalin was93%.The yield of amygdalin was over 60% and the purity was more than96% after single factor experiment was carried out.Enrichment and purification of mother liquor: In order to improve the total yield of amygdalin,the mother liquor which contains about 30%of amygdalin was enriched and refined.Through macroporous resin enrichment and recrystallization,the yield of amygdalin in mother liquor can reach more than 18%,and the total yield of amygdalin can reach more than 78%,and purity of amygdalin was more than 96%.The pilot-scale study: In order to verify the feasibility of this process,we made two pilot scale-up test(10 kilograms class and 100 kilograms class).The yield of amygdalin reached over 75% and the purity reached more than 96%,which indicates that the process is simple and suitable for industrial production.Quality evaluation of amygdalin: The quality evaluation of amygdalin,which was obtained from small scale and pilot-scale,was studied.The quality evaluation includes characters,melting point,loss on drying and purity.And the stability of amygdalin was studied,which provides reference for the quality standard and preservation condition of amygdalin.
