The Discovery Of Terpenoids And Research On Bioactivities Of Dolomiaea Souliei (Franch.) Shih And Caesalpinia Minax Hance

Dolomiaea souliei?Franch.?Shih,belonging to the Dolomiaea genus of Compositae,is mainly distributed in Sichuan Province,China.Its roots have been used as a traditional Chinese herbal medicine to alleviate pain in abdominal distension and stomach diseases.It is commonly used for the treatment of gastrointestinal diseases.Caesalpinia minax Hance,belonging to the Caesalpinia genus of Leguminosae,is widely distributed throughout tropical and subtropical regions of Southeast Asia,also in China's Guangdong,Guangxi,Yunnan,Guizhou and Hainan Province.Its seed,named as‘‘ku-shi-lian'',has been used in traditional folk Chinese medicine for the treatment of common cold,dysentery,rheumatism,wound and poisonous snake bites,and so on.This thesis made a comprehensive chemical components research,respectively,on the Dolomiaea souliei of Compositae and Caesalpinia minax of Leguminosae.Various chromatography methods,such as silica gel column chromatography?CC?,preparation thin layer chromatographic?PTLC?,thin layer chromatographic?TLC?,Sephadex LH-20 and recrystallization were used to separate the 41 compounds.With a variety of spectral analysis means,including optical rotation,UV,IR,EI-MS,ESI-MS,NMR(¹H-NMR,¹³C-NMR,DEPT,¹H-¹H COSY,HSQC,HMBC,NOESY),and X-ray single crystal diffraction experiment were applyed to identify all isolated compounds.The ethanol extract of the roots of D.souliei were separated by various methods to get 21 known compounds,including eleven sesquiterpene lactones,three triterpenes,one anthraquinone,one steroid,two furan derivatives and three other compounds.Among them,two compounds were the first separated from the family Compositae and two compounds were first isolated from Dolomiaea genus,also two were separated from this plant for the first time.The ethanol extract of the C.minax were isloated to get 20 compounds,including thirteen diterpenes,one triterpene,one flavone,one steroid,one furan derivative and three other compounds.Among them,two are new compounds.New compounds CM-01 and CM-02 were tested for biological activity against Alzheimer's disease?AD?using C.elegans CL4176 paralysis experiment method.Test data shows that compound CM-02 have moderate activity against AD and compound CM-01 have no activity.Finally,we have summarized variety resources,chemical compositions and biological activities of the Dolomiaea genus.We want to provide the reference value for better development and utilization of the medicinal value of the Dolomiaea genus and discover more features and activities compounds.
