medicinal-preparation of Liantai Fang based on drug systems;The fifth chapter:summary and discussion.Chapter 1:summary of the related literaturesSummaring the history and current situation of research progresses in the chemical constituents and pharmacological research of Liantai Fang,118 pieces of references were cited.Summaring the current situation of Huanglian Jiedu Decoction.26 pieces of references were cited.Summaring the current situation of the cerebral ischema reperfusion injury model and the basic research of traditionl Chinese compound material.43 pieces of references were cited.Chapter 2:studies on the medicinal-preparation technology of the effective substance of Liantai Fang based on drug systemsThis paper was based on the thought that natural pharmacy concept drug system was naturalness,cooperativity and affinity,which was a link between pharmacokinetic and pharmacodynamics(PK-PD-DI).Through studying Liantai Fang,we found that total flavonoids,total alkaloids and total phenols components in Liantai Fang were the effective components based on drug systems.Total alkaloid and total flavonoids extraction ration and transferring ratio of the total alkaloids and total flavonoids of the effective parts were the indicators,studying the preparation technology of purifying the total flavonoids,total alkaloids and total phenols of Liantai Fang.Liantai Fans was comprised of Huanglian Jiedu Decoction and cinnamon.Firstly,based on preparation technology 1 and 2,we got the corresponding sediment b1 and b2 from the effective component of Huanglian Jiedu Decoction.We put the extract liquid in P,type on macroporous adsorption resin(diameter ratio was 1:H1),the sample amount content and resin volume ratio of 1:V1,dynamic adsorption the sample with G1BV water after elution,and we used G2BV E1%ethanol elution,elution velocity G3ml/min,collected the E1%alcohol eluent,recycled solvent,vacuum drying.We sot the corresponding enrichment c1 and c2 from effective component of Huanglian Jiedu Decoction.The results of the purification technology validation showed that the content of the total flavonoids and total alkaloid content of the effective substance of Huanglian Jiedu Decoctionl and the effective substance of Huanglian Jiedu Decoction 2 was stable.The content of total flavonoids of Liantai Fang is 10.12%.The extract ratio of total flavonoids was 88.66%,94.98%respectively.The extract,enrichment of total content of flavonoids of Huanglian Jiedu Decoction 1 was 14.92%,21.02%,16.95%.The extract,sediment,enrichment of total content of flavonoids of Huanglian Jiedu Decoction 2 was 18.22%,26.83%,12.15%.The transferring rate of total flavonoids of sediment from Huanglian Jiedu Decoctionl and Huanglian Jiedu Decoction 2 was 38.96%,26.24%.The transferring rate of total flavonoids of enrichment was 36.68%,26.78%.The extract ratio of total alkaloids was 93.94%,69.50%respectively.The content of extract,sediment,enrichment fo total content of alkaloids of Huanglian Jiedu Decoction 1 was 35.64%,43.57%,42.21%.The extract,sediment,enrichment of total content of alkaloids of Huanglian Jiedu Decoction 2 was 30.06%,47.42%,27.34%.The transferring rate of total alkaloids of sediment from Huanglian Jiedu Decoction 1 and Huanglian Jiedu Decoction 2 was 35.82%,40.51%.The transferring rate of total alkaloids of enrichment was 26.30%,26.73%.The results of the purification technology validation showed that the extract ratio of index component Baicalin from Huanglian Jiedu Decoction 1 and Huanglian Jiedu Decoction 2 was92.09%,89.94%respectively.The content of Baicalin of the extract,sediment,enrichment from Huanglian Jiedu Decoctionl was 8.55%,16.27%,8.12%.The content of Baicalin of the extract,sediment,enrichment from Huanglian Jiedu DecoctionZ was 9.52%,17.28%,7.68%.The transferring rate of Baicalin of sediment from Huanglian Jiedu Decoctionl and Huanglian Jiedu Decoction 2 was 54.67%,30.63%.The transferring rate of Baicalin of enrichment from Huanglian Jiedu Decoctionl and Huanglian Jiedu Decoction2 was 31.85%,30,68%respectively.The extract ratio of index component Berberine from Huanglian Jiedu Decoctionl and Huanglian Jiedu Decoction 2 was 90.12%,89.41%respectively.The content of Berberine of the extract,sediment,enrichment from Huanglian Jiedu Decoctionlwas 9.09%,14.89%,9.71%.The content of Berberine of the extract,sediment,enrichment from Huanglian Jiedu Decoction2 was 10.28%,14.84%,11.33%.The transferring rate of Berberine of sediment from Huanglian Jiedu Decoctionl and Huanglian Jiedu Decoction 2 was 46.05%,35.06%.The transferring rate of Berberine of enrichment from Huanglian Jiedu Decoction 1 and Huanglian Jiedu Decoction 2 was 24.21%,41.66%respectively.Secondly,we researched the best extraction process of total phenolic from cinnamon and made a comprehensive analysis on total phenol extraction quantity index,studying the extraction technology of cinnamon phenolics.Using L9(43)orthogonal experiment design,the extraction of total phenol amount as an index,examined the extraction solvent(A),extraction times(B),extraction time(C)and solvent ratio(D).Combined with the actual production,we determined the best extraction technology was A3B3C2D1,namely A%ethanol reflux,extraction B3 times,extract hours C2,dosage of solvent for D1 times,and we got the preparation of cinnamon extracts.We measured the right amount of cinnamon,added 10 times the amount of water soaking 1 h and water vapor dstillation for 8 h.After that we got the essential oil from the cinnamon.3 batches of samples of extraction technology showed:the average extraction ratio of the cinnamon essential oil was 2.57%,the average extraction rate of the extract wasl 1.6%.Chapter 3:studies on compatibility of different medicinal-preparation of Liantai Fang based on drug systemsThe effective substance 1 compatibility pharmacodynamic studies had shown that Huanglian Jiedu Decoction(b1+c1)preparation technology was the best,reflecting the preparation of process design and optimization of the effectiveness of the precision of its preparation.The experimental results of SOD,MDA showed that the compatibility of components Huanglian Jiedu Decoction 1 could obviously increase the SOD activity and reduce MDA content of model rats.The preparation of the effective substance of Huanglian Jiedu Decoction 1 group(b1+c1)and group(b2+c2)Huanglian Jiedu Decoctionl could decrease MDA content,had the extremely significant difference(compared with model group,P<0.01).The cinnamon effective substance compatibility pharmacodynamics research had shown that cinnamon extract and volatile oil compatibility group had significant difference compared with model group,but other components of cinnamon effect was not obvious.The experimental results of SOD,MDA showed that compared with model group,each component of cinnamon could obviously increase the MDA content of model rats and lower SOD levels compared with model group(P<0.01,P<0.05),which damage free radicals caused by ischemia.lt showed that each material component of cinnamon could not alleviate cerebral ischemia caused by free radical damage.The effective substance compatibility pharmacodynamics research of Liantai Fang showed that the best preparation technology of Liantai Fang was group(b1+c1),the optimum combination group was Liantai Fang combination 1(b1+c1+d+e).This combination had improved neurologic symptoms and against cerebral ischemia in rats.Liantai Fang combination(b1+c1+d+e)could significantly improve the model rats neurological symptoms compared with model group(P<0.01)and Liantai Fang combination group had significant difference compared with model group.Chapter 4:quality representation of the medicinal-preparation of Liantai Fang based on drug systemsEpicatechin reference substance solution and cinnamon color after a sample solution were at 768±2nm wavelength with maximum absorption,thus wavelength at 768 nrn was confirmed.Ferric chloride,potassium ferricyanide method was developed for the determination of total phenolic chromogenic method was:precisely absorbing a certain amount of sample solution,put it in a 25 mL volumetric brown flask,adding 70%methanol to 5 mL,respectively.Add 2.0 mL of 0.3%sodium dodecyl sulfate solution and 0.6%ferric trichloride-0.9%potassium ferricyanide mixture(1:1)2.8 mL.Maintain it in the dark place for 8 minutes,then the absorbance values at 768nm wavelength was confirmed.We established methods for the deternination of the content of the total phenoland and we used the high performance liquid chromatography(HPLC)method for the determination of Baicalin as the index component of the total flavonoids of Huanglian Jiedu Decoction and Berberine as the index component of total alkaloids of Huanglian Jiedu Decoction.Geniposide,Cinnamic acid,Cinnamic aldehyde,Phellodendrine,Berberine hydrochloride as the index component of Liantai Fang.The quantitative method of total cinnamon phenolics content was established by using UV detector.HPLC-DAD method was used in the experiment to determine the effective components in the medicinal-preparation of Liantai Fang.The linear regression of Geniposide was Y=918893x+175129,r2=0.9994(n=6),the favourable linear relationship in the range of 0.2?g?0.7?g.The linear regression of Phellodendrine was Y=452352x+8931.68,r2=0.9990(n=6),the favourable linear relationship in the range of 0.0052?g?0.065?g;the linear regression of Berberine paper summarized the work and discussed the basis of Huanglian Jiedu decoction compound research,clarified the the rationality and validity of Liantai Fang and cinnamon compatibility.The innovation points were as follows:1 The quantitative method of total cinnamon phenolics content was established by using UV detector.2 The high performance liquid chromatography(HPLC)method for the determination of Baicalin,Geniposide,Berberine Hydrochloride,Cinnamicacid,Cinnamic aldehyde,was established.3 The optimal extraction process of total phenol of cinnamon was established.The best medicinal-preparation technology of Liantai Fang and the optimum combination group was determined.
