The experimental study of Capparis spinosa selenium polysaccharide on human hepat oma HepG2 cells cell growth inhibition and apoptosis,and discuss its pathways and m echanisms.In this paper,we investigate the Capparis spinosa selenium polysaccharide on human hepatoma HepG2 cell growth inhibition by MTT and inverted microscope method.Exp erimental results show that when the concentration of in the wild watermelon selenium polysaccharides become increasing,inhibition rates of cell growth will rise Capparis spin osa selenium polysaccharide detecting the role of HepG2 cells was observed by inverted microscope,cells were suspended.And the higher the concentration of Capparis spinosa selenium polysaccharide,the longer action time and the phenomenon is more obvious.Observing the fluorescence microscopy showed that the increasing concentrations of Cap paris spinosa selenium polysaccharide increases produced more the number of apoptotic b odies.We observed different concentrations of Capparis spinosa selenium polysaccharide t o the role of apoptosis in HepG2 cells using flow PI staining method.The results sho w that with the increasing of the concentration of Capparis spinosa selenium polysacchari de,apoptosis rate also increased.In confocal microscopy,Capparis spinosa selenium polys aecharide effect HepG2 cells after 48h,about the change of reactive oxygen species?RoS?level,the concentration of Ca2+ and mitochondrial membrane potential.The result s show that with the increasing of the concentration of Capparis spinosa Selenium Polysacc haride,elevated the levels of ROS,the concentration of Ca2+,decline the value of mito chondrial membrane potential.The results indicate,Capparis spinosa Selenium Polysaccharide can cause the apoptosis of human hepatoma HepG2 cells and increase the levels of ROS.Because of excessive ROS,mitochondrial inner membrane will be damaged,resulting the calcium influx in cell cytoplasm.ROS elevated concentrations of calcium ions within the mitochondria,w hich further damaged the mitochondria,resulting decreased of mitochondrial membrane potentialin.As result,releasing of apoptosis-related factors induce apoptosis.In sumnary,this thesis demonstrated Capparis spinosa Selenium Polysaccharide can induce human hepatoma HepG2 cell apoptosis by in vitro experiments and mitochondr ial pathway is Capparis spinosa Selenium Polysaccharide induces apoptosis in human he patoma HepG2 cell apoptosis may be one way.
