Objective:This study was on the basis of rat models of post stroke depression,Through pathology,Behavioral test,immunohistochemistry,RT-PCR mRNA detection and FGF-2 compared by inject of NBP drug intervention.Preliminary discussion the FGF-2'role in depression after stroke mechanism of nerve cells and the protective effects of Butylphthalide injection.Method:1.Preparing PSD rat model and experimental groups:prepare several healthy male Wistar rats which it's weight between 250g-300g.All experimental animal model of(OPEN-FIELD method)were selected by OPEN-FIELD method before surgery.The animals who have similar scores were chose,obtaining 56 rats.The qualified rats(56)were divided into 4 groups randomly:sham operation group(n=8),the stroke group(n=8),PSD model group(n=20),drug intervention group(n=20)after weighing and sorting.Establish rat model of PSD by permanent bilateral common carotid artery ligation(2-VO),followed by solitary curing method and chronic unpredictable mild stress.Drug intervention group received butylphthalide by gastric perfusion,20mg/kg/d on the second day after surgery;the rats of PSD model group and stroke group,sham operation group were given the equal dose of saline.According to the observation and execution time of,the four groups were randomly divided into the group of two days after modeling(2D);nine days group(9d);sixteen days group(16d);twenty-three days group(23d)and four sub group.Sham operation group and stroke group were each 2 animals.PSD group and drug intervention group were each 5 animals.2.Behavioral test of the PSD rats:Behavioral test of the PSD rats:depression level was evaluated by spontaneous activities,interests,self exploration abilities of the rat through Open Field Test(OFT);compared the weight changes among groups;detecting interest and pleasure of rat by sugar consumption.3.Then use immunohistochemistry technique to probe the FGF-2 'expression level of the dynamic changes in the nerve cells.Use fluorescence quantitative-PCR to detect FGF-2 changes in mRNA expression in the cells.Through the observation of dynamic changes in different sub groups to describe the observed indexes.Results:1.Praxiology Test shows:?Weight ' s changes of each group:Compared with the Sham operation group,after modeling,There was a slow increase of the rats' weight,or even declined in the Stroke group?PSD-group?NBP-group,and it was obvious from the 9 th day after modeling(P<0.05);Compared with the Stroke group,the NBP group have no significant difference at each time point,P>0.05;Compared with the PSD group,the rats' weight of the NBP-group growth in the point 16d?23d,with statistical significance(P<0.05).?Sucrose water consumption test:The consumption of each group decreased from the second day after modeling,the stroke group(P<0.05)?the PSD group(P<0.01)?the NBP group(P<0.01)were lower than the shan operation group;Compared with the stroke group,the PSD group had a significant difference in the 16' th day(P<0.05)and the 23 ' th day(P<0.05);Compared with the PSD group,there were more consumption of the NBP group in the 9' th?16 'th?23 ' th day after modeling(P<0.05).?Open Field Test:Compared with the sham operation group,the activity level of the stroke group(P<0.05),PSD group(P<0.05)and NBP grouP(P<0.05)decreased after modeling,especially in PSD group;On the points of the 16' th day and the 23' th,there were significant differences between the NBP group and the PSD group(/P<0.05).The NBP can improve rats' s activity.Compared with the sham operation group,the vertical ability of the PSD group declined with the change of time(P<0.01);The NBP group's vertical exploration ability decreased in the 9 days after modeling,then gradually recovered,it was more significantly improved than the PSD group's.The vertical exploration ability of rats improved after NBP applicated.2.Immunohistochemistry:Compared with the sham operation group,the expression of FGF-2 of the stroke group's was higher with a statistical significance(p<0.05)in the 9' th day and the 16' th;Compared with the stroke group,the PSD group,in the 9' th?16 ' th and the 23 ' th day after modeling,had a significant difference(P<0.05);Compared with the stroke group,the NBP group had a significant difference(P<0.05)in the 9' th?16' th and the 23 ' th day after modeling.3.RT-PCR:The FGF-2 mRNA expression in the PSD group showed a downward trend,it was significant(P<0.05)in the 9d?16d?23d,to the PSD group;The highest expression of FGF-2 mRNA come in the 7' th day from the NBP intervented after modeling(P<0.05),which was match with the immunohistochemistry results?Conclusion:1.The intervention of NBP-injection can promote the expression of FGF-2 and improve the PSD rats' activity,interest and exploring ability effectively.
