| Objective To observe the effect of modified Hungfeng decoction on proteinuria a nd dynamic changes of expression of podocin in rats with IgA nephropathy,and to fu ther explore the mechanism of proteinuria of IgAN and try to find the approach to th e therapy for IgAN through modeling IgAN rats.Also it aims to provide more scienti fic and adequate theoretical basis for assessing efficacy of traditional Chinese medicine therapy to IgA nephropathy.Methods Rat IgAN model was modeled by the improved method of oral feeding of bovine serum albumin(BSA)+carbon tetrachloride(CCL4)hypodermic injection+lipopolysaccharide(LSP)vena caudalis injection.SD rats were randomly divided into normal group(N),IgAN model group(M),controlled group of tripterygium(T),modified Hungfeng decoction low-dose group(L),modified Hungfeng decoction high-dose group(H).The controlled groups was given tripterygium,treatment groups are were given decoction of modified Hungfeng at the finishing of replicating model for 8 weeks.N group and M group were given normal saline.Quantitative urinary protein of 24 hours,blood urea nitrogen,serum creatinine,serum albumin were teseted.The IgA intensity was examined by direct immunofluorescence.The pathological changes of renal tissue HE stained were observed by light microscope.With a transmission electron microscope to observe the glomerular podocyte.The expression and distribution of podocin in renal tissue was examined by immunohistochemistry and Western Blot.The mRNA expression of podocin in renal tissue was examined by Real time-PCR.Results(1)Comparing with the normal group,the UPEs in other groups were significantly increased(p<0.05).At the 17h weekend,comparing with the model group,the UPEs in three administration groups were significantly decreased(p<0.05).It had no significant difference in UPEs in three administration groups(p>0.05).(2)At the 17th weekend,it had no significant difference in rats of serum creatinine,blood urea nitrogen and serum albumin in each group(P>0.05).(3)The fluorescence intensity of IgA in N group ranged from-?±.IgA deposition looked like branches thick and coarse granular in mesangial areas in M group,and the fluorescence intensity of IgA ranged from++?+.Other groups of IgA deposition in mesangial areas were significantly reduced(P<0.05).(4)M group has manifestation of mildly and moderately mesangial matrix and cell proliferation.T group,L group,H group were improved compared with M group.(5)M group has most of glomerular foot process fusion,some foot process disappears,or part of microvilli appear,mesangial area electron dense deposits seen,there are significant differences compared with the N group.Each administration group showed foot process fusion reduced and partially visible normal foot process architecture.(6)Protein levels showed that podocin protein in M group,T group,L group,H group were significantly decreased compared to N group(P<0.05).Podocin protein in T group,L group,H group were significantly increased compared to M group(P<0.05),no significant difference was showned among the three groups(P>0.05).(7)Expression of mRNA showed that the podocin mRNA of renal tissue in M group was markedly lower than N group(P<0.05).The expression of podocin mRNA in T group,L group,H group were higher compared to M group(P<0.05).There was no statistical difference between the three administration group(P>0.05).Conclusion Modified Hungfeng decoction has the function of decreasing urinary protein,inhibiting mesangial cells and mesangial matrix proliferation,decrease the deposition of IgA in rats with IgAN model,decrease the foot process.It raises the expression of podocin protein and mRNA in rats.It hints that modified Hungfeng decoction can reduce urinary protein excretion by raise the expression of podocin protein by regulating the expression of podocin,reduce the injury to podocyte and recover the damage of glomerular filtration membrane barrier. |
