Objective(1)To observe the effects of Buyanghuanwu Decoction(BYHWD)promotes a ngiogenesis on the migration of neural stem cells,and to explore the underlying mech anism after Cerebral Ischemia.Methods Mice were subjected to middle cerebral artery occlusion(MCAO)for 30 min utes.BYHWD(20 g/kg)was administered orally 24 h after ischemia,once daily for 14 days.The and the corner test were used to evaluate sensorimotor on 1?7 and 14 days after ischemia respectively.Fourteen days after ischemia,the expression of vWF and DCX was detected by immunofluorescence,the expression of CXCR4?SDF-1 and BDNF mRNA was determined by real time PCR,and the expression of CXCR4 prot ein was examined by western blot.Results(1)Compared with the control group,BYHWD significantly ameliorated neurol ogical dysfunction and reduced the number of right turn at 7 and 14 days after ische mia(P<0.05).(2)BYHWD can significantly increased the expression of vWF and DC X(P<0.01),however,given endostatin reduced the expression of vWF and DCX in t he ischemic cortex(P<0.01).Within this neurovascular niche,newly born,immature n eurons closely associate with the remodeling vasculature.(3)Compared with the contr ol group,BYHWD significantly increased CXCR4?SDF-1 and BDNF mRNA expressi on and BDNF?CXCR4 protein expression in ischemic cortex(P<0.01).Conclusion(1)BYHWD accelerates functional recovery after cerebral ischemic.(2)Treat ment with BYHWD induced angiogenesis and neuroblast migration along a blood vess el scaffold toward the post-stroke cortex.(3)SDF-1/CXCR4 and BDNF expression ind uced by BYHWD treatment promotes neuroblast micro-vascular coupling after stroke.
