Endophytic bacteria is the mainstream of the field of microbial research and an important source of new antibacterial activity material screening.Endophytes isolated from shield leaf yam,identified as bacillus licheniformis,it not only has the ability of saponin production,but also can secrete large amounts of extracellular products,and have antibacterial activity.This study on the strain cremation cultivation and shaking flask fermentation liquid,through the system of organic solvent extraction separation of antimicrobial active substances,in order to the separation and purification,find out a new antibacterial lead compounds.Through experimental study,the following results:(1)Compared to PDA fermented liquid and LB fermented liquid antibacterial rate found that LB fermented liquid is generally better than PDA fermented liquid antibacterial activity.(2)The fermented liquid extract diluted 50 times the test strains of inhibitory effect is not obvious,and the inhibition rate of individual strains were negative.(3)Through the study of the activity determination of organic solvent extraction phase,a)strains of petroleum ether extraction were provided by the relative inhibition difference is more apparent,inhibition rate of corn bending spore pathogen is most obvious,the inhibition rate increased as the concentration of petroleum ether extraction phase as;But for individual 53 species,3 pathogenic bacteria such as wheat,rice blast bacteria inhibition rate is negative,are proved under the concentration,relative to the petroleum ether extraction were strains without inhibition,on the contrary,in promoting the growth of the selected strains.Ethyl acetate extract b)five selected strains were provided by the relative inhibition,ethyl acetate extract relative corn bending spore pathogens inhibition rate without petroleum ether extraction phase inhibition rate obviously,but the ethyl acetate extract relative apple anthrax bacteria has obvious inhibitory effect,the inhibition rate increase with the concentration of the ethyl acetate extract phase increases as well.C)relative eight kinds of n-butanol extraction test fungi all have different degrees of inhibitory effect,but the difference is bigger.Similarly,n-butyl alcohol extract relative test fungi inhibition rate is increased with the increase of concentration.D)was associated with significantly lower inhibition rate of methanol extraction phase,only 3 pathogenic bacteria of wheat and apple anthrax bacteria have higher inhibition rate;Second,the inhibition rate is not present increases with the increase of the concentration and bacteriostatic activity of methanol extraction phase containing less material.(4)By thin layer chromatographic separation and n-butyl alcohol extraction,separation of the five components,and calculate the corresponding yield.(5)By GC-MS analysis of petroleum ether extraction phase,get 40 kinds of components,can be divided into esters,alcohols,acids,aldehydes,ketones,hydrocarbon and sulfur,chlorine and organic matter.Including esters content accounted for 59.05%,hydrocarbon accounted for 22.68%,other several types of the material content is less.The tributyl phosphate ester is the first time in microbial metabolites of light component,found that 91.36%of the ester matter.Second,there is a small amount of the light component 3-isobutyl six hydrogen pyrrole and[1,2-a]pyrazine-1,4-diketone,mainly by means of chemical synthesis of such compounds,in the light component of microorganism metabolites have been found,the compounds have good medicinal value.(6)Antibacterial activity test in the field,in order to shield yam leaves endogenous bacillus licheniformis SYt1 fermented supernatant on diluted 50 times as potions,northwest agriculture and forestry university of science and technology college of horticulture grape resource base infected with grapes black blain pathogen vine(jade,Beijing,zhengzhou early Sp522)do foliar spraying test,test showed that shield yam leaves of bacillus licheniformis fermented supernatant on diluted 50 times with no obvious inhibitory effect.
