Study On The Standard Extract Of Chuanxiong Volatile Oil

Chuanxiong is the dried rhizome of Ligusticum wallichii Hort for the Umbelliferae plant,with promoting blood circulation of Qi,dispelling wind and relieving pain.The volatile oil(mainly in ligustilide)is the main active ingredients in Chuanxiong.Chuanxiong has many producing areas and this makes its quality mixed.According to the preliminary research,the main active ingredients in Chuanxiong volatile oil are lactones,which are unstable that can be changed during extracting,separating and storing,which makes its quality reduced.Besides,there is no quality standard for Chuanxiong volatile oil in Chinese Pharmacopoeia yet,and seriously affect its application in the field of clinical medicine and pharmacy.So,it's urgnt to study and make strict quality standard for the standardized extract of Chuanxiong volatile oil.The work as follows:1.Taking the extraction rat of Ligustilide as evaluation index,we adopt orthogonal experiment to optimize the best extraction process of steam distillation for Chuanxiong volatile oil.The results show that the best extraction conditions were 6 times volume of water and 1.5%Ethyl Acetate in oil-water separator for 10 hours.2.By using the medium-pressure column chromatography to separate the volatile oil and identified it by HPLC and NMR,4 ingredients were identified,which were 2-methoxy-4-vinylphenol,senkyunolide A,Z-ligustilide and 3-Butylidenephthalide.3.The contents of ligustilide and relative content changes of senkyunolide A,3-n-Butylphthalide and 3-Butylidenephthalide at different times were detected by HPLC when the volatile oil were respectively put in refrigeration(4?)avoid light,room temperature(18?25 ?)avoid light and room temperature under light.As a Result,the degradation rate constants of ligustilide in volatile oil put in the three conditions were 2.0×10-4,6.4×10-3 and 8.4×10-3;the relative content of 3-n-Butylphthalide and 3-Butylidenephthalide elevated while ligustilide and senkyunolide A decreased,and the increased range of 3-Butylidenephthalide was close to the reduction of ligustilide.The result of stability shows that Chuanxiong volatile oil should be stored below 4? and avoid light.4.The research on quality standards of Chuanxiong volatile oil was including Character,identification,inspection and determination.Using Psoralen as an internal standard and the method of multi-components quantitation by one marker,the determination of ligustilide and total lactones were detected by HPLC,among which,the total lactones included senkyunolide A,3-n-Butylphthalide,ligustilide and 3-Butylidenephthalide.
