Purpose:Chronic bronchitis is a threat to public health worldwide, and it is a major clinical features with recurrent cough, sputum or accompanied by wheezing common respiratory diseases.Chronic bronchitis is a common disease in our country, the high incidence. In recent years, with the aging of the population and the degree of air pollution increases the incidence of chronic bronchitis is increasing, seriously affecting the ability to work and quality of life, the World Health Organization(WHO)as one of the research the disease. In this paper, through the danxiankechuanning capsules anti-inflammatory effects and TNF-?,IL-1? effects of inflammatory mediators and IL-6 in the NF-?B signaling pathway in chronic bronchitis of rats,a more systematic discussion danxiankechuanning capsule drug to treat chronic bronchitis possible molecular mechanism for the treatment of chronic bronchitis developed to provide experimental evidence.Method:1 Using xylene-induced ear edema, egg white induced paw edema induced granuloma Law and cotton research guts cents Kechuanning capsules anti-inflammatory effects.2 Smoked method using modified copy model of chronic bronchitis. The modeling of the rats were placed in a special chamber 1m3 of smoke with wood shavings, sawdust, tobacco fumes ignited each 50g,2 times a day, every 30min,sustained 40d.Blank control group were placed in a smoke-free environment of normal feeding.3 The 48 SD rats of either sex were randomly divided into six groups, each 8.Note:control group(Control),model group(Model),bile cents Kechuanning high dose group(High),bile cents Kechuanning capsule dose group(Middle),bile cents lower Kechuanning Capsules dose group(Low),positive control group(DXM).Using oral gavage route of administration, and in the end after modeling the second day of administration.Blank control group and model control group were given normal saline, once a day for 21d.4 Were observed and recorded the general state of the rats before and after modeling and before and after administration, including mental status, activity, breathing, coat color and body weight change.5 Rats were killed after thoracotomy a complete separation of the lung tissue.lung tissue morphology naked eye.HE staining, light microscopy of lung tissue morphology.6 Was observed by immunohistochemistry in lung tissue of rats with chronic bronchitis NF-?B(p65)expression.7 By enzyme-linked immunosorbent assay(ELISA)in serum and lung tissue of rats with chronic bronchitis and inflammatory mediators TNF-a,IL-1?,and the content of IL-6.Results:An anti-inflammatory effect results danxiankechuanning capsules showed that compared with the control group, the bile cents Kechuanning capsule could inhibit the ear swelling in mice, significantly inhibited paw edema in rats, significantly inhibited due tampons granulation tissue proliferation.2 Improved fumed copy chronic bronchitis model results show that the model rats with prolonged smoked time, there are different degrees of coughing and breathing deeply, mouth nasal color deepens, there are obvious secretions, less tired lying move, mental fatigue, reduced feeding, brown hair gradually dry.No significant differences in body weight of rats in each group before the experiment, declined after the end of the experiment modeling smoked rats weighing each group compared with the control group of rats body weight was significantly(P<0.01);after administration, danxiankechuanning capsules higher, dose group and positive control group gradually improved mental state, increased hair gloss, breathing gradually stabilized, oral nasal secretions decrease, increase activity levels, diet increases.Weight model control group than the control group significantly decreased body weight, cholesterol danxiankechuanning capsules high dose group and positive control group it was significantly higher than the model group(P<0.01).3 Bronchopulmonary morphological observation results showed that after administration, visually gall cents Kechuanning capsules of high, medium and low dose group and positive control group part of the lung tissue color dark red, occasionally spotting, compared with the model group there are significant differences.Light microscopy gall cents Kechuanning high dose group and positive control group showed bronchial epithelial cells intact,no obvious wall thickening, bronchial lumen without significant expansion, with see a small amount of inflammatory cell infiltration;dose group showed bronchial epithelial cells partially complete, partial expansion of the bronchial lumen, associated with chronic inflammatory cell infiltration.Low-dose group showed bronchial epithelium compared with incomplete lumen without significant expansion, inflammatory cell infiltration.4 Immunohistochemistry showed that compared with the model group, gall cents Kechuanning capsule high, medium and low dose group and positive control group of rats can reduce lung tissue NF-?B expression(P<0.01 or P<0.05)·5 Enzyme-linked immunosorbent assay(ELISA)showed that, compared with the model group, gall cents Kechuanning capsule high, medium and low dose group and positive control group can reduce serum and lung tissue TNF-a,IL-1? and content(P<0.01 or P<0.05)IL-6 's.Conclusion:1.By xylene-induced ear edema, egg white induced paw edema and cotton-induced granuloma method shows guts cents Kechuanning capsule has significant anti-inflammatory effect.2 By the general state of rats with chronic bronchitis, changes in body weight and lung tissue morphology, indicating fumed copy chronic bronchitis model reliability.3 By inhibiting NF-?B(p65)expression,reduce inflammatory mediators TNF-a,IL-1(3,and IL-6 levels, the process NF-?B signaling pathway and its inhibition has some relevance.
