The Study Of The Screening Technique For ShuangHuangLian Injection Allergen Chip

Objective:The method of this experiment was adopted by combining immunity bottle chip and immunity cover chip together with ELISA as its basic principles.It provided a preliminary groundwork for the preparation of Shuanghuanglian injection allergen chip.Methods:This screening of allergenic compounds was carried out by making small sample chip from shuanghuanglian injection.First,sensitize the laboratory animal by injecting holoantigen which was formed by BSA and Baicalin,chlorogenic acid?baicalein?violaguercitrin?Ursolic acid?Chrysin from Shuanghuanglian injection,respectively.Then,separate and purify the blood serum after sensitization is confirmed.One portion of the treated blood serum was injected into laboratory animal to form antiantibody,the other portion was used for enzyme labelled antibody by IgG enzyme marker.Based on the crossed ELISA reaction,screening of allergen component from ShuangHuangLian injection was carried out by using both immunity bottle chip and immunity cover chip together.Results:The immunity bottle chip and immunity cover chip analysis results indicate that allergenic ingredients in Shuanghuanglian injection are not baicalein?violaguercitrin?Ursolic acid?Chrysin.But baicalin,chlorogenic acid is the most suspicious sensitized composition in ShuangHuangLian injection.Conclusion:In this research,small sample chip was produced successfully by combining immunity bottle chip and immunity cover chip together due to crossed reaction,which is used for screening sensitinogen in Shuanghuanglian injection.This system can be well used for screening sensitinogen in the Chinese materia medica Injection in the future?Its application can not only provide references for clinical drug safety,but also can pave the way to establish a method for high-flux analyses of allover ingredients in Chinese materia medica and their Formula.
