Liposarcoma is a malignant mesenchymal tumor derived from fat cells and the mesenchymal cells differentiation into adipocyte.It can occur in each position of the whole body,but see mainly after deep soft tissue of lower limb and peritonaeum.It divides into on histology well differentiated,dedifferentiated,myxoid,round cell,and pleomorphic five shape types liposarcoma.Due to the high rate of recurrence and metastasis in liposarcoma and its polymorphous cause diagnostic confusion,the correct diagnosis and treatment is particularly important in clinical work.AFAP1L1 is a member of the AFAP family along with AFAP1.The actin-filament associated protein have been shown can impact actin filament crosslinking,active cSrc and participate in the actin cytoskeleton arrangement.In the current study,we investigated AFAP1L1 and AFAP1 expression in adipose tissue tumor using immunohistochemical and in situ hybridization.We analyze the relationship between the actin-filament associated protein and clinical significance in liposarcoma to provide new biological indicator for clinical diagnosis and prognosis judgment.Materials and Methods1 Materials:Test to collect 62 cases,including 27 ALN/WDL,14 MLS,19 DL and 2 PLS.Additionally,to collect 20 lipomas.2 Methods:Max Vision immunohistochemical staining methods and in situ hybridization were used to detect the expression of AFAP1L1 and AFAP1 in liposarcoma and control group.Results1 The expression rates of AFAP1L1 and AFAP1 protein in liposarcoma were 71.0%(44/62)and 40.3%(25/62),and the control groups were 15%(3/20)and 5%(1/20).The expression pattern between LPS and other control groups has statistical difference(P<0.01).2 The expression rates of AFAP1L1 and AFAP1 mRNA in liposarcoma were 40.3%(25/62)and 29%(18/62),and the control groups was all negative.The expression pattern between LPS and other control groups has statistical difference(P<0.01).The expressions of AFAP1L1 and AFAP1 on the protein level were in coincidence with that on the mRNA level.3 The expression pattern between AFAP1L1 and AFAP1 protein in LPS has statistical difference(P<0.05).Spearman correlation analysis showed they were negative correlation(r=-0.343,P<0.05).4 The expression of AFAP1L1 in LPS was obviously associated with recurrence or metastasis(P<0.01),but not with tumor sites(P>0.05),location(P>0.05)and differentiation degree(P>0.05).Kaplan-Meier survival analysis shows that the survival rate of positive AFAP1L1 is lower than that of the negative group.By the Log-rank test the expression pattern between positive and negative has statistical significance(? 2=8.128,P<0.05).The positive rate of AFAP1 has no relation with various clinical parameters.ConclusionThe expression of AFAP1L1 and AFAP1 in LPS may play an important role in the occurrerce and progression of LPS.AFAP1L1 shows close relation to the recurrence and metastasis in LPS,may be a novel prognostic factor and one of the novel targets for LPS.
