The Effect Of Realgar On Mice Liver Toxicity

Realgar is a commonly used mineral medicine,it is very important in Chinese traditional medicines.Realgar has detoxification,insecticide,dampness,phlegm,malaria and other functions in traditional medicine,to do more for the external use.While in modern medicine realgar has antibacterial,antiviral,anti-tumor effects,as the medication use.But the type of insoluble rough mineral and an overdose of cinnabar in drugs,have led to toxicity exceeding international criterion.Meanwhile,oral Realgar often makes somebody uncomfortable.So it and realgar-containing medicine are restrict in international market.So this research used Kunming mice as experimental animals,studied on realgar inducing liver injury by intragastric administration,and tried to reveal the mechanism of realgar inducing liver injury,for scientific and rational use of realgar providing a theoretical basis.Realgar dose were 1/32LD50:0.1g/kg;1/16LD50:0.2g/kg;1/8LD50:0.4g/kg in experiment.The research determined the effect of Realgar on mice liver weight and organ coefficient and made histological observation,the effect of Realgar for mice serum alanine aminotransferase(ALT),aspartate aminotransferase(AST),alkaline phosphatase(ALP)activity and y-glutamyl transpeptidase(y-GT)activity,and the effect of Realgar on mice liver cell cycle and liver cell membrane ATP enzyme activity.The experimental results showed that realgar could inhibit the increase of body weight in mice compared with the blank group,liver coefficient increased significantly.Histological observation showed realgar could cause swelling,degeneration,necrosis and proliferationof mice liver cell.Activity of mouse serum ALT low,middle dose group increased comparing with the blank group,but there was no statistical significance,high dose group increased significantly,activity of AST,ALP and ?-GT,in low middle and high dose group than in the control group had obvious increasing trend,this explained Realgar had certain damage effect on mice liver.There search of Realgar on mice liver cell cycle indicated that the increase of the dose could arouse retardant of the cell cycle,cell function and liver cell injury;At the end activity of mice liver cell membrane ATPase in each dose group was significantly reduced compared with the control group.The above results indicate that Realgar can cause serious liver injury in mice,and make activity of liver ATPase decreased,resulting in insufficient energy,the ultimate cause of severe hepatic injury.In summary,after long-term administration of Realgar on mice weight plays a restraining effect,on liver tissue and the function of liver produces certain effect,and can injury liver cells,making activity of the liver ATPase decreased,ultimately resulting in severe hepatic injury.
