Cycloastragenol?CAG?is the sapogenin of most astragalosides,which can be prepared from astragaloside IV?ASI?.It has been reported that CAG is an effective anti-aging ingredient by activating the telomerase and retarding shorten the telomere length.In this article,the extraction process of CAG has been optimized with the indicator of CAG content.Based on the above studies,components contained in the CAG extract were identified tentatively by LC-MS.In addition,biological activity of the CAG extract has been studied.Details are as follows:1.The process of preparation of CAG ectract was optimized with the indicator of CAG content.The optimum pocess is as follow:Crude powder of Radix Astragali was refluxed with 75%alkaline ethanol?pH12?for 2h twice.And then the extract was purified by macroporous resin chromatography,polar impurities were removed by water and 30%of ethanol.After that,70%of ethanol was used to get ASI enrichment product.Then the extract was dissolved in 20%methanol-water solution,oxidized with one equivalent NaIO4 for 12h,then one equivalent NaBH4was added for 4h reduction reaction.Finally,acidified with 1 M H2SO4 at pH 1.5-2.0 for 24 h.The temperature was controled at 35?.2.The extract was subjected to a component identification.A total of35 compounds were identified in positive and negative mode through Thermo LTQ Orbitrap XLMTT system.It provides a theoretical basis to the pharmacological activity research of the CAG extract.3.H2O2 was used in PC12 to establish an aging mode.MTT assay was used to evaluate the protective effect to aging cell of CAG.The results indicated that CAG concertration range in 0.4?M-2?M showed a significant protective effect,which indicate the anti-aging effect of CAG.It fits in former studies in literature.4.Based on the aging mode established previously,the biological activity of CAG extract was determined by MTT method,age-related beta-galactose glucoside enzyme method and ELISA method.Results showed that the ectract has obvious telomerase citivation,but indistinctive anti-aging effect.
