The functional mechanism of the general anesthetics has been the important topics in research.Gas signal molecule widely participate in anesthetics mechanism and play important roles in the process.Hydrogen sulfide as a new type of gas signal molecule and CBS is the only enzyme to produce it in the central nervous system.We still don't know whether they take part in the process of anesthesia.In this content,three kinds of typical general anesthetics were chosen to investigate the influence on H2S and CBS in the central nervous system on rats.The objective of the study is to investigate the relations between H2S,CBS and anaesthesia.240 wistar rats were randomly divided into saline solution control group(CG),isoflurane group(IG),ketamine group(KG)and zoletil group(ZG).Each group except CG was sub-divided different subgroups:anesthesia subgroup,anesthesia recovery subgroup I and anesthesia recovery subgroup ?.Rats in CG were injected saline,IG were inhalation isoflurane,KG were injected ketamine and ZG were injected zoletil.Rats were decapitated in relative time,then hippocampus,brainstem,thalamus,cerebral cortex and cerebellum tissue were separated.The content of H2S,expression area and protein of CBS were determined by a variety of methods.Experimental results such as follows1.H2S from different brain regions was measured by spectrophotometrically with methylene blue.During the rats anesthetized,the content of H2S decreased with the deepening of anaesthesia induction,and increased when recovery from anesthesia in hippocampus,brainstem and cerebral cortex(P<0.05);While there wasn't significant changes in thalamus and cerebellum(P>0.05).2.Immunohistochemical staining were carried out to study the CBS expressed in each brain regions.It revealed that regional expression in hippocampus while decentralized expression in other brain regions.3.Western blotting was used to detect protein expression of CBS.During the rats anesthetized,the expression of protein CBS decreased with the deepening of anaesthesia induction,then return to normal levels when recovery from anesthesia in hippocampus and brainstem(P<0.05).The protein expression of CBS in cerebral cortex differ between general anesthestics.The protein expression of CBS in thalamus and cerellum has no significant difference(P>0.05).Conclusion:Three general anesthetics during anesthesia changes in concentrations of H2S in the brain and CBS is consistent with the behavioral changes in rats,indicates that general anesthetics mechanism with H2S and CBS content in the brain may exist a certain correlation,namely the H2S and CBS may participate in the body of general anesthetics activities related regulation in the brain.
