ObjectiveTo observe the effects mechanism of hydrogen sulfide on the osteoporosis in Ovariectomized Rats, and provide experimental basis for prevention and treatment of osteoporosis in postmenopausal osteoporosis.MethodsThe SPF level of healthy female SD rats in age of 6 monthes were selected and randomly divided into 5 groups: sham operation group, model control group, hydrogen sulfide high, medium and low dose group. Sham rats were suffered bilateral ovarian exploratory surgery. Other rats were underwent bilateral ovariectomy, resulting to ovariectomized rat model. The hydrogen sulfide group rats were treated with different doses of exogenous H2 S donor GYY4137(10, 30 and 50mg/kg). 12 weeks later, rats were sacrificed and the blood samples were collected for measurement of Ca2+, P and E2, and alkaliphosphatase(ALP), osteocalcin(OC) and tartrate-resistant acid phosphonatase(TRACP) in the serum. Real-time PCR and Western blot were detected in the expression of OPG and RANKL.Results(1) 4 weeks after ovariectomy, the concentration of Ca2+, P and E2 in serum significantly decreased. After GYY4137 treatment, the level of Ca2+, P and E2 increased gradually.(2) The content of ALP and OC was decreased, and the TRACP was increased in the OVX group, as compared than that in the SHAM group. GYY4137 treatment could increase the level of ALP and OC, but decrease the TRACP in serum.(3) The mRNA and protein level of OPG in OVX group were decreased, as compared with the SHAM rats. While they were significantly increased after GYY4137 treatment.(4) The m RNA and protein of RANKL were increased. After treatment of GYY4137,it became increased, as compared the OVX group.Conclusion(1) H2 S could increase the concentration of Ca2+, P and E2 in serum and improve the bone turnover marker in Ovariectomized Rats(2) H2 S could decrease the level of RANKL and increase the ratio of OPG/RANKL.
