| Guangdong Herbal(produced by limited company of Guangzhou Wang Laoji)is brown granules,and is mainly composed of the extract of ilex asprella,helicteres angustifolia,lophatherum gracile,oroxylum indicum,microcos,polygonum chinense,desmodium styracifolium,rosa laevigata,lygpdium japoncum and citrus medica.It tastes sweet and bitter,and was widely used for prevention and treatment of flu,fever,sore throat and other disease.However,there are very few literatures reported so far,meanwhile,witth the successing to apply World Cultural Heritage,so it is important to explain the mechanism and principle by scientific methods.1.The results of the preliminary test of Guangdong Herbal:the chemical constitutnets of Guangdong Herbal mainly contains flavonoids,polyhpenols,saccharides,saponins,terpenoids,lactone and coumarone,such as may contain tannins,anthraquinone,organic acids,and it laies a foundation to further research the chemical constitutents and active ingredients.2.Evaluate the quality of Guangdong Herbal by the TLC fingerprint:(1)the different polars parts extracted by different polarity solvent contains kinds of chemical constituents;(2)it shows that the Guangdong Herbal has a wide distribution polarity through different solvent system results;(3)the TLC fingerprint results related to the adsorption materials;(4)the TLC fingerprint results are consistent from different batches,showing that the stable qulity of Guangdong Herbal;3.Research on the chemical constituents of Guangdong Herbal:the Guangdong Herbal was extracted methanol,then partitioned with petroleum ether,ethyl acetate and n-BuOH sequentially,especially.The EtOAc and n-BuOH extractions were subjected to the column chromatography over silica gel,ODS,Sephadex LH-20 or HPLC.As a result,twenty-four compounds were obtained and their structures of twenty-two compounds were identified on the basis of physicochemical properties and spectral analysis,and named as:ilexasosides I(1),ilexside B,asprellanosides B(3),asprellanoside A(4),2a,3a,19a-trihydroxyursa-12-en-28-oic acid ?-D-glucopyranosyl ester(5),2a,3?,19a-trihy-droxyursa-12-en-28-oic acid ?-D-glucopyranosyl ester(6),2?,3?,19?,23-tetrahydroxyurs-12-en-28-oic acid ?-D-glucopyranosyl ester(7),4-p-cumaroyl-D-glucose(8),chlorogenic acid(9),6-hydroxy-4?-(4-hydroxy-3-methoxyphenol)-3 a-hydroxyl-meth1-5-methoxy-3,4-2-naphthaldehyde(10),caffeic acid(11),E-p-coumatic acid(12),isoviolanthin(13),4',5,7-trihydroxyflavone-6-O-?-D-glucopyranosyl ester(14),6-C-glycopyranosyl luteolin(15),(+)-catechin(16),3-O-methy-lellagic acid 4'-O-rhamnopyranoside(17),protocatechuic acid methyl ester(18),protocatechuric acid(19),gallic acid(20),p-hydroxybenzoic acid(21),sucrose(22).Among them compound 1 is a new compound,and is a sulfuric acid ester based ursa contains alkanes type monosaccharide triterpenoid glycosides compounds,8 is firstly obtained as a natural product;1-10,12-15,17-18 compounds were first isolated from the Guangdong Herbal,also from isolated particles triterpene compounds.4.Identifying of frigerprint information research of Guangdong Herbal:According to information of the chemical constitutents,the major ninety-five peaks of the established Base Peak Chromatography were detected,and sixteen peaks has been assigned.In addition,the chmical structure of the eightyth peak was inferred by the TOF-MS and IF-MS. |
