Herba Justicia,also named as Xiaoqingcao,Sijiaoying and etc.,derives from the whole dried plant of Rostellularia procumbens(L.)Ness(Acanthaceae).Due to the effects of clearing heat and detoxifying,diuresis detumescence,preventing malaria,promoting blood circulation and relieving pain,it is often used in clinic practice for treating the cold and fevers,malaria,sore throat,infantile malnutrition,diarrhea,nephritis edema,urinary system infection,etc.,and topically treating carbuncle furuncle,traumatic injury.Distributed widely in South China,southwest of Taiwan province and other places,this plant tends to grow in a damp environment,and is available from wilderness in late summer or autumn,fresh or dried for medication,as recorded in the Chinese Pharmacopoeia of 1977 edition.Modern pharmacological studies have confirmed the brilliant aggregation activity of medicinal Justicia,of which the main sort of active ingredients have been investigated as lignans and concentrated in ethyl acetate fraction through our preliminary experiments.In this study,four fractions incluing petroleum ether,ethyl acetate,n-butanol,aqueous solution were separated from the 80%ethanol extract of Justicia by the method of system solvent.Using optical nephelometry on a platelet aggregation analyzer,with ADP as inducer,80%ethanol extract and these four fractions were put into test successively,the result showed that the ethyl acetate fraction has a most significant anti-platelet aggregation effect withaIC500.120mg/mL,whichrevealedapotentiality for Justicia to be developed into an innovative anti-platelet aggregation medicine.On the other side,HPLC-DAD and fluorescence analysis were taken as guides for detecting object compounds lignans,21of which were isolated by means of several chromatographic processes as silica column,Sephadex LH-20 gel column,Toyopearl HW-40F gel column,preparative TLC and preparative HPLC chromatography,from the ethyl acetate extraction.14 compounds’structures were elucidated by magnetic resonance spectroscopy(NMR)method or mass spectrometry(MS),including 7 lignans:neojusticin A,Justicidin E,neojusticin B,Chinensinaphthol mether ether,JusticidinB,cilinaphthalide B,6′-hydroxy justicidin B,and 7 lignanosides:Justicidinoside C,procumbenoside B,Cleistanthin B,Justicidinoside B,procumbenoside A,Justicidinoside A,Tuberculatin.Among the last unrevealed 7 lignanosides,probably at least 2 of them were obtained from Justicia procumbens or Acanthaceae plants for the first time.This study laid a material foundation for the subsequent pharmacodynamics and mechanism study of anti-platelet aggregation activity of Justicia.
