Objective:To investigate the different metabolic index and its related genes SNPs and to analyze the association of the different metabolic index with its related genes SNPs in Korean-Chinese and Han nationality pupils of Yanbian area.Methods:1206 elementary school students(706 of Korean-Chinese and 500 of Han nationality)were selected in 3 of primary schools from August,2011 and August,2013.Using epidemiological investigation methods combined with laboratory measurement,1)the general information were collected by the standardized questionnaire;2)the body mass index(BMI),waist circumference(WC),systolic blood pressure(SBP)and diastolic blood pressure(DBP)were measured;3)blood triglyceride(TG),high-density lipoprotein cholesterol(HDL-C),glucose(FBG)and insulin(FIns)were measured after 12 hour fasting;4)DNA was extracted with the Genomic DNA Kit provided by TIANGEN BIOTECH(BEIJING)Company Limited;5)genotypes of NOS3 gene rs3918227 site,PCK 1 SNPs gene rs 1042531 site and ADIPOQ gene rs 17846872 site were detected by using Sequenom MassARRAY(?)SNP combined with multiplex PCR,single base extension and matrix-assisted laser desorption/ionization-time of flight technics;6)human IR was estimated by the homeostasis model assessment method(Homeostasis model assessment,HOMA)Results:1.Distribution characteristics of genes SNPs that’s related with the different metabolic index 1)The CC,CA and AA genotype frequencies at the NOS3 gene SNP rs3918227 site were 93.5%,6.2%and 0.3%in Korean-Chinese nationality pupils and 84.4%,15.6%and 0.0%in Han nationality pupils,respectively,and nationality difference was observed for genotype frequencies(P<0.001);the C and A allele frequencies were 96.6%and 3.4%in Korean-Chinese nationality pupils and 84.4%and 15.6%in Han nationality pupils,respectively,and there was significant difference in the distribution of allele frequencies between two nationality pupils(P<0.001).2)The TT,GT and GG genotype frequencies at the PCK1 gene SNP rs1042531 site were 59.5%,36.0%and 4.5%in Korean-Chinese nationality pupils and 61.6%,33.2%and 5.2%in Han nationality pupils,respectively,and there is no nationality difference was observed for genotype frequencies(P=0.567);the T and G allele frequencies were 73.4%and 26.6%in Korean-Chinese nationality pupils and 61.6%and 38.4%in Han nationality pupils,respectively,and there was significant difference in the distribution of allele frequencies between two nationality pupils(P<0.001).3)The GG,GT and TT genotype frequencies at the ADIPOQ gene SNP rs 17846872 site were 90.1%,9.3%and 0.6%in Korean-Chinese nationality pupils and 86.0%,14.0%and 0.0%in Han nationality pupils,respectively,and nationality difference was observed for genotype frequencies(P=0.011);the G and T allele frequencies were 94.8%and 5.2%in Korean-Chinese nationality pupils and 86.0%and 14.0%in Han nationality pupils,respectively,and there was significant difference in the distribution of allele frequencies between two nationality pupils(P<0.001).2.Uni-variate analysis for the association of the different metabolic index with its related genes SNPs 1)In the different genotype of NOS3 gene SNP rs3918227 site,there were different distribution of WC,SBP,DBP and HOMA-IR in Korean-Chinese nationality pupils(P<0.05)and there were different distribution of BMI and FBG in Han nationality pupils(P<0.01);in the different allele frequencies,there was different distribution of WC in Korean-Chinese nationality pupils(P<0.05)and there were different distribution of BMI and FBG in Han nationality pupils(P<0.001).2)In the different genotype of PCK1 gene SNP rs1042531 site,there were different distribution of BMI,WC,SBP,DBP and FBG in Han nationality pupils(P<0.01);In the different allele frequencies of PCK1 gene SNP rs1042531 site,there were different distribution of BMI,WC,SBP,DBP and FBG in Han nationality pupils(P<0.05)and there were different distribution of HDLC in Han nationality pupils(P<0.05).3)In the different genotype of ADIPOQ gene SNP rs17846872 site,there were different distribution of WC,DBP and TG in Korean-Chinese nationality pupils(P<0.05)and there were different distribution of TG and HOMA-IR in Han nationality pupils(P<0.05);in the different allele frequencies,there was different distribution of WC in Korean-Chinese nationality pupils(P<0.05)and there were different distribution of BMI,TG and HOMA-IR in Han nationality pupils(P<0.05).3.Multivariate Logistic regression analysis for the association of the different metabolic index with its related genes SNPs The multivariate Logistic regression analysis showed that:1)the different allele at the NOS3 gene SNP rs3918227 site was closely related with nationality,BMI,WC and FBG,and the value of OR was 5.710(95%CI:4.013-8.132),1.195(95%CI:1.097-1.302),0.939(95%CI:0.908-0.971)and 1.537(95%CI:1.124-2.102),respectively;2)the different allele at the PCK1 gene SNP rs 1042531 site was closely related with nationality,DBP,TG and HDLC,and the value of OR was 2.209(95%CI:1.831~2.664),1.010(95%CI:1.002~1.019),0.851(95%CI:0.718~1.009),and 0.638(95%CI:0.444~0.917),respectively;3)the different allele at the ADIPOQ gene SNP rs17846872 site was closely related with nationality,BMI,DBP,TG,FBG and HOMA-IR,and the value of OR was 3.382(95%CI:2.472~4.626),1.060(95%CI:1.007~1.116),0.987(95%CI:0.9 72~1.001),1.668(95%CI:1.369~2.032),respectively.Conclusion:1)There are SNPs of NOS3 gene rs3918227 site,PCK1 gene rs1042531 site and ADIPOQ gene rs 17846872 site in Korean-Chinese and Han nationality pupils of Yanbian area.2)NOS3 gene rs3918227 and ADIPOQ gene rs17846872 are significant differences for the distribution of genotype and allele frequencies at them between two nationality pupils;PCK1 gene rs1042531 is significant differences for the distribution of allele frequencies between two nationality pupils.3)There is different association of the different metabolic index with its related genes SNPs and there are nationality differences.
