Objective:To investigate the expression levels of OVCA1mRNA in epithelial ovarian tumorsand to explore molecular mechanism in the development ovarian cancer,to set up theoretical basis for diagnosis and therapy of epithelial ovarian cancer.Methods:Collectionovarian tissues 28 cases form resection,including 8 cases of normal ovarian tissues,7 cases of benign ovarian tumortissues,13 cases of epithelial ovarian cancertissues,extract total RNA fromRNAiso plus method,after agarosegel electr-ophoresis and ultraviolet spectrophotometer,determine RNA purity and integrity.quantitative real-time PCR detected the expression levels of OVCAlmRNA.Statistical analysis was performed using SPSS 20.0 software.Receiver operating characterristic(ROC)analysis was performed to further evaluate the clinical value of OVCA1mRNA expression for the prediction of epithelial ovarian cancer.Results:1.After electrophoresis total RNA,observed two bands,the fluorescence intensity of 28S is greater than 18S,measurement absorbance ratio of A260/A280 are 1.8 to 2.0,suggests that RNA purity and integritymeet the requirements.2.There was higher level of OVCA1mRNA expression in epithelial ovarian cancer group(15.81±5.44)than in normal ovarian tissue group(1.70±0.43)and benign ovarian tumor group(0.90±0.63)(P<0.05).There was no significant difference of the expression levelsbetween normal ovarian group(1.70±0.43)and benign ovarian tumor group(0.90±0.63)(P>0.05).3.ROC analysis of theexpression OVCA1mRNA levels in ovarian cancer,area under the curve(AUC)is 0.897.95%credibility interval confidence interval is 0.767-1.0,according to blind method inferred,sensitivity is 76%,specificity is 87%,so OVCAlmRNA could beconsidered as a candidate marker for the diagnosis of ovarian cancer.Conclusions:1.There was high level ofOVCA1mRNA expression in epithelial ovarian cancer.2.OVCA1mRNA could be a candidate marker for the diagnosis of epithelial ovarian cancer.
