Research About MicroRNAs In Plasma And Peripheral Blood Mononuclear Cell In The Diagnosis Of Schizophrenia

Abstract/Summary:
Objective: This study explored the differential expression of the plasma and peripheral blood mononuclear cells(PBMCs)microRNA(miRNA)in patients with schizophrenia and normal controls by researching on the expression profiles and levels of the plasma and PBMC miRNA in patients and healthy controls.Characteristic diagnostic models of miRNA in peripheral blood for schizophrenia were created.The diagnostic value of these models in Chinese patients with schizophrenia was estimated.Method: 1).We screened out 7 miRNA(has-mir-449 a,has-mir-34a-5p,has-mir-652-3p,has-mir-564,has-mir-432-5p,has-mir-548d-5p and has-mir-572)according to the literature.Anticoagulant whole blood samples from 35 schizophrenia patients were collected as disease group.Anticoagulant whole blood samples from 15 healthy subjects visiting our physical examination center were selected as control group.The plasma and PBMC were separated in 2 hours after phlebotomy and they were saved in an environment of-80 degrees Celsius immediately.2)We got primer sequences of the seven miRNAs through query research and database.Then we commissioned AiQi Biotechnology Company(Sichuan,China)to synthesis,purify and identify the primers.3).RNAs were extracted from plasma and PBMC.Real-time RT-PCR was used to detect the types and expression levels of miRNAs from both schizophrenia patients and healthy control.4).The experimental results were collected and analyzed.5).Thediagnosis model was established by statistical analysis of the experimental data.And its efficiency was validated and tested.Results: Four of the seven miRNAs in disease group from both plasma and PBMC were up-regulated and the difference was statistically significant(P<0.05).The expression of the remaining three miRNAs has no obvious gap(P>0.05).The united diagnosis models were established for schizophrenia by the four differentially expressed miRNAs.The area under ROC curve of plasma was 98% and 86% of PBMC.The expression levels of the seven miRNAs were over-expressed in plasma than in PBMC and the difference was statistically significant(P<0.01).Conclusion: From what has been discussed above,miRNAs may be used for the diagnosis of schizophrenia.Because of the source of plasma and PBMC is convenient and miRNAs are stable in plasma and PBMC,miRNAs in plasma and PBMC may be new blood biomarkers for the diagnosis of schizophrenia.
