Objective Schizophrenia is a complex genetic disease which usually result in a shortened lifespan. As the impairment in the patient's learning or working ability,it brings heavy burden to the affected, the families and the society.Biomarkers that accurately reflect the pathophysiology of schizophrenia contribute to the diagnosis, treatment and prognosis.Micro RNAs are small non-coding RNA transcripts constructed with single-stranded.Studies have found that micro RNAs can regulate the expression of special genes or proteins at the post transcriptional level, which disrupt neurobiological processes, especially in schizophrenia.Through literature review, this thesis investigates the differential expression of micro RNA-30 e in the plasma of patients with schizophrenia and health controls by means of quantitative Reverse Transcriptase Polymerase Chain Reactions(q RT-PCR).The correlation analysis will also be performed between the expression level of micro RNA-30 e and the positive and negative symptom scale(PANSS),personal and social functioning scale(PSP)of the first-episode schizophrenia.Method The plasma from 17 first-episode patients and 16 health controls both aged 17-45 years without sex-limited were collected for the detection of micro RNA-30 e.The PANSS and PSP were also performed in the schizophrenia subjects.Result1.Compared with the health, the plasma level of micro RNA-30 e in the patients with schizophrenia was decreased significantly(p<0.05).2.In the schizophrenia, there were not significantly correlation between the expression levels of micro RNA-30 e in plasma and the whole scores of PANSS,positive symptoms, negative symptoms, general psychopathology symptoms(p>0.05);the Partial correlation coefficient between the expression levels of micro RNA-30 e and the scores of aggression risk was at 0.517,the difference was statistically significant(r=0.517,p=0.040).3.There were not significantly correlation between the expression levels of micro RNA-30 e in plasma of schizophrenia and the whole scores of PSP,beneficial activity of the society, personal and social relationships,self-care,interference behavior(p>0.05).Conclusion1.The dysregulation of micro RNA-30 e in plasma may be a potential biomarker for the diagnostic of schizophrenia.2.The expression levels of micro RNA-30 e in schizophrenia has no significant correlation with the scorces of PANSS or the scale of PSP,but the Impulse control ability, suggesting micro RNA-30 e could be trait rather than state-dependent marker.
