Objective:To study the relationship between vitamin D receptor gene Fok I polymorphism and the severity of tuberculosis in sichuan han population.Methods:Case-control study was conducted in this study.Under the principle of informed consent,the whole blood and serum of tuberculosis patients and health adults in sichuan han nationality adults were collected.61 tuberculosis patients(34 cases of severe patients,27 mild patients,ages 18 to 60 years old)were enrolled in study group,49 health adults(age 18 to 60 years old)who has no sibship with each other were enrolled in control group.Serum VDR concentration was measured by ELISA(enzyme-linked immunosorbent method),and Fok ? genotypes were detected by PCR-RFLP(polymerase chain reaction-restriction fragment length polymorphism).Hardy Weinberg law was used to verify the groups' representative;Analyzing the differences of SNP(single nucleotide polymorphisms)genotypes and allele distribution between these groups;Analyzing the relationship between three genotypes and the serum VDR concentration.Data were analyzed by SPSS 20.0 statistical software.Results:1.The groups were conformed to the law of Hardy-Weinberg.2.?The distribution frequencies of genotypes between Tuberculosis(TB)group and control group of VDR-FF,Ff,ff were 36.1%,54.1%,9.8%and 28.6%,51%,20.4%,respectively,P>0.05.There was no statistically significant difference.?The distribution frequencies of genotypes between TB severe group and mild group of VDR-FF,Ff,ff were 41.2%,47.1%,11.8%and 29.6%,63%,7.4%,respectively,P>0.05,and there was no statistically significant difference.3.The relationship between genotypes and serum VDR concentration:the serum VDR concentration of FF group,Ff group,ff group was 8.01 ± 1.57 ng/ml,6.48 ± 1.69 ng/ml,5.26 ± 1.64ng/ml,F=17.924,P<0.001,and the significant differences were found.4.?The serum VDR concentration of TB group was 8.018 ± 1.4 ng/ml,while the control group was 5.29 ± 1.08 ng/ml,t=11.271,P<0.001,and the significant difference was found.?The differences of serum VDR concentration were found between three groups,p<0.05.Conclusion:1.VDR Fok I polymorphism may have no obvious correlation with the susceptibility and severity of tuberculosis in sichuan han population.2.In sichuan han population,The level of serum VDR concentration of VDR-F allelic gene may be higher than that of f allelic gene.3.In sichuan han population,the level of serum VDR concentration may have correlation with the incidence of tuberculosis;In adult patients with tuberculosis,the level of serum VDRconcentration may have positive correlation with the severity of tuberculosis.
