Effect Of Jitongning Capsule On Rat With Rheumatoid Arthrits And Its Mechanism Reaearch

Objective:To observe the therapeutic effect of Jitongning capsule(JTN)on rats with rheumatoid arthritis(RA).From the level of cell research Jitongning capsule on R A mechanism of the imbalance of Th17/Treg cells.Methods:Except for the normal group,on the basis of the collagen induced arthritis(CIA)in rats,using wind,cold,wet environment factors" copy the wind-cold-dampness arthralgia of arthritis in Wistar rats.Model rats were randomized into 5 groups:the model control group,the Tripterygium Glycosides(TG)group,the JTN rather low,medium and high dose group.The body weight,articular swelling degree and the arthritis index(AI)were measured once a week.HE method to detect the articular histopathological changes and evaluate the degree of rat ankle joint damage with the pathology score.To weigh of rats with spleen and thymus,calculate the coefficient of spleen and thymus coefficient.Flow cytometry to detect the spleen of Th17 and Treg cells.ELISA method to detect the rat inflammatory cytokines in serum TNF-? and the levels of IL-6,Western Blot method to detect the synovial membrane the expression of JAK1,STAT1 and STAT3 protein levels.Results:JTN decreased the articular swelling rate,foot swelling degree and AI,and relieve inflammation of the joints on body weight,alleviate the pathological damage,reduce pathological scores.JTN significantly decreased spleen coefficient and thymus coefficient.JTN increased significantly spleen Th17 accounted for the proportion of CD4+T cells and total lymphocyte.Medium and high dose group of Treg accounted for a significant rise in the proportion of CD4+ T cells,high dose group of Treg significantly rose proportion of total lymphocyte,Th 17/Treg cells percentage in JTN decreased obviously.rather low,medium and high dose group of rats serum TNF-? and IL-6 levels significantly declined.The synovial tissue of rats JAK1,STAT1 and STAT3 protein positive expression of stripe is abate,with the increasing dose,the expression of stripe.Conclusion:JTN improve RA multiple symptoms of arthritis,relieve joint swelling and inhibit the synovial inflammatory cell infiltration and cartilage injury.JTN activate Treg cells,inhibit Th 17 cell proliferation and activation,the trend of Th 17/Treg balance,ease the development of joint inflammation.JTN reduce rats serum TNF-? and IL-6 level,and to a certain degree of inhibition of the activation of JAK1,STAT1 and STAT3,probably by blocking the J A K/STAT signal pathway,thus inhibiting the production of inflammatory cytokines,protection of rheumatoid arthritis rats.
