Object:To investigate the important effector molecules of Huangqiliuyi decoction acting on Type 2 Diabetes Melhtus(T2DM)by integrating network analysis at a system level.Then the treatment effects of Huangqiliuyi decoction on the treatment of T2DM were explored through the analysis and research of pharmacology experiment and transcriptomics.Methods:The main effective elements in the treatment of Huangqiliuyi decoction in T2DM were predicted by using network pharmacology method.Seventy-two male diabetic rats were randomly divided into blank control group,model group,positive group,and Huangqiliuyi decoction(high,medium and low dose group),including high,medium and low dose group doses were crude drug 12.60g/kg,6.30g/kg,3.15g/kg,irrigation gastric administration,once a day,for four consecutive weeks.Measurement of body weight and blood glucose changes every week.The transeriptome of pancreas from these rats were sequencing by using Illumina sequencing platform.Based on the predict target,the gene expression levels of the blank group,model group and Huangqiliuyi decoction high dose group were calculated.Results:Firstly,According to the prediction of network pharmacology,20 major candidate effector molecules of Huangqiliuyi decoction on T2DM therapy were discovered.Secondly,Huangqiliuyi decoction can significantly increase the body weight of T2DM rats,while reduce the fasting blood glucose and glycated hemoglobin.The result showed that the hypoglycemic effect of Huangqiliuyi decoction had posstitive correlation with the dosage.Thirdly,the transcriptome of pancreas were sequencing by using Illumina sequencing platform.On the basis of eggNOG,gene expression could be classified into the following significant expression patterns:the unknown functional class(24.25%),which Is the largest classification unit;the follow are energy production and conversion,Amino acid transport and metabolism?Nucleotide transport and metabolism,Carbohydrate transport and metabolism,Coenzyme transport and metabolism,Lipid transport and metabolism.Fourthly,by analyzing the results of Illumina sequencing and network pharmacology prediction,Huangqiliuyi decoction high dose group and model group were compared.GO analysis of up-regulated genes included NAD-dependent protein deacetylase sirtuin-1,Androgen receptor,Nuclear factor NF-kappa-B p105subunit,Insulin receptor substrate-1,Adenosine receptor,Solute carrier family 2 facilitated glucose transporter member 4,Polyubiquitin-C,cysteinyl aspartate specific Proteinase-3,Glucocorticoid receptor;The down-regulated genes included Epidermal growth factor receptor,Transcription factor p65,Protein kinase C beta type,Insulin receptor substrate-2,cysteinyl aspartate specific proteinase-8,Histone deacetylase,Amyloid beta A4 protein,Tumor necrosis factor receptor superfamily member 1A,Tumor necrosis factor?Conclusion:Huangqiliuyi decoction can significantly reduce fasting blood glucose,glycosylated hemoglobin in T2DM rats.The predicted results of network pharmacology of Huangqiliuyi decoction Treatment,together with the transcriptome sequencing results suggested that Huangqiliuyi decoction can regulate Transcription factor p65,NAD-dependent protein deacetylase sirtuin-1,Insulin receptor substrate-2,Androgen receptor,Adenosine receptor,Solute carrier family 2 facilitated glucose transporter member 4,Glucocorticoid receptor,Tumor necrosis factor,Histone deacetylase,Protein kinase C beta type,Tumor necrosis factor receptor superfamily member 1 A,while did not effective function the Epidermal growth factor receptor,Polyubiquitin-C,Nuclear factor NF-kappa-B p105subunit,cysteinyl aspartate specific proteinase-3,cysteinyl aspartate specific proteinase-8 and the molecular mechanism of Insulin receptor substrate-1 is unclear.Because the subjects used in this experiment are the male rats,the results not contain Progesterone receptor and Estrogen receptor expression data.
