Objective:To develop the compound health food particles Z,which can enhance the immunity.Method and Results:?1?In order to set the prescription which can improve the immunity,low mice immunized with different characteristics on the impact of hemolysin antibody generation and the DNFB induced mice skin delayed-type hypersensitivity?DTH?test were studied.The experimental results of hemolysis and DTH show that JL4 prescription namely this study group prescription can enhance immunity function.?2?Adopt the single factor investigation and orthogonal experiments,with the ganoderma lucidum polysaccharide transfer rate and the dry paste yield for the comprehensive evaluation index to select the best extraction.Finally optimize the best extraction of ganoderma based on L9?34?orthogonal experiment was:20 times of water is used to extract three times,1 hour each time.?3?Single factor test was used to screening the type and amount of materials.With the hygroscopicity,formablity,liquidity and dissolubility as the comprehensive evaluation indexs to optimize the best molding process.Through the research of molding process,finally choose dextrin as auxiliary materials,taste masking agent choose aspartame,75%alcohol as the wetting agent,12mesh granulating,60?dry,14 mesh whole grins.?4?TLC identification methods were used for ganoderma lucidum,A herbs and Notoginseng.The TLC results show that the ganoderma lucidum?A herbs and notoginseng had chear TLC spots,good specificity,and the negative control without interference.Using anthrone-sulfuric acid spectrophotometric method to detect the content of ganoderma lucidum polysaccharide,the results of content determination show that the ganoderma lucidum polysaccharide in 0.00250.015 mg·mL-1concentration range had good linear relationship?r=0.9997?,the average recovery of 98.5%,RSD was 1.6%;Using HPLC to detect the content of gallic acid?the total content of panax notoginseng saponins R1?ginseng saponin Rg1?Re?Rb1 and Rd in compound health food particles Z.The resule of Gallic acid in the range 0.01120.3371mg.mL-1 had a good linearity?r=0.9998?,the average recovery of98.3%,RSD was 2.7%.Notoginseng saponin R1 in 10.26256.5 m g·?L-1 and ginseng saponin Rg1 in 34.21855.2 m g·?L-1?Re in3.69192.26 m g·?L-1?Rb1 in 29.14728.6 m g·?L-1and Rd in6.132153.3 m g·?L11 concentration range respectively into good linear relationships?r=1.000,r=0.9999,r=0.9999,r=0.9997,r=0.9999?,the average recovery for 95.9%?98.8%?98.5%?100.8%?99.6%,with RSD 2.9%?2.8%?1.5%?2.1%?2.7%.?5?Using the commercially available plastic composite film packaging materials packaging the compound health food particles Z for the 3 months of preliminary stability test,results show that the character,content,granularity of compound health food particles Z examining indexes up to specification.Conclusion:This study result show that the preparation technology was reasonable and feasible.The TLC had chear spots,the methods had good specificity and sensitivity.The content determination methods had good repeatability and accuracyt,easy to operate.The methods can be used for the qualitative and quantitative identification of the compound health food particles;Experiment of health products granules quality is stable and controllable.
