Objective : By detecting the levels of Atopobium cluster and Collinsella aerof-aciens in normal glucose tolerance(NGT)and type 2 diabetes mellitus(T2DM)in Uygurs and Kazaks of Xinjiang,analyze the correlation between Atopobium cluster,Collinsella aerofaciens and type 2 diabetes.Methods :Collect NGT and T2 DM blood and fecal samples in Uygurs and Kazaks of Xinjiang,using 16 S r DNA real-tim-e PCR technology to detect the levels of Atopobium cluster and Collinsella aerofa-ciens in fecal samples.The correlation analysis between Atopobium cluster,Collinella aerofciens,and FBG,height,weight,BMI,TC,TG,HDL-C,LDL-C were conducted by us-ing Pearson.Resluts:1.Compared NGT in Kazak population,the level of Atopobium cluster,Collinsella aerofaciens in T2 DM group in Kazaks showed significant differe-nce(P=0.012,P =0.024).There was significant different between the quantity of Atopobium cluster,Collinsella aerofaciens in NGT of the two national(P=0.025,P=0.007).2.Atopobium cluster was positively correlated with the level of HDL-C(r=0.281,P=0.031),and it was negative correlated with the levels of weight(r=-0.692,P=0.001),BMI(r=-0.400,P=0.002),TC(r=-0.503,P=0.000),LDL-C(r=-0.581,P=0.000);the level of Collinsella aerofaciens was positively correlated with the levels of HDL-C(r=0.550,P=0.000),and it was negative correlated with the levels of weight(r=-0.449,P=0.000),BMI(r=-0.432,P=0.001).Conclusion:Atopobium cluster and Collinsella aerofaciens may be related to lipid metabolism of Type 2 diabetes mellitus in Uygur and Kaza-k.Objective:With gatifloxacin as a tool intervent normal glucose tolerance and in-sulin resistance in mice,to observed the changes of blood biochemistry,intestinal C.-coccoides subgroup,Desulfovibrio and Plasma amino acid,analyze the correlation be-tween C.coccoides subgroup,Desulfovibrio,amino acids and insulin resistance.Metho-ds:1.Using high fat diet to establish insulin resistance in mice model of 16 weeks,the insulin resistance mice were randomly divided into two groups by glucose,lipids and body weight,model group and Gatifloxacin administration group(120mg/kg/d).according to the same batch of normal tolerance mice were randomly divided into normal glucose tolerance group and treatment group(Gatifloxacin 120mg/kg/d)with normal glucose tolerance,gavage five weeks,blood lipid and glucose were measure b-y glucose oxidase method and enzymatic(COD-PAP,GPO-PAP);Enzyme-linked immu-noassay detection fasting plasma insulin levels in mice,and calculate the insulin res-istance index,insulin sensitive index;16S r DNA real time PCR to detect levels of C.coccoides subgroup,Desulfovibrio in fecal samples in mice;The levels of plasma a mino acids were detected by HPLC.2.Using Pearson analysis,multiple stepwise regre ssion analysis and path analysis,to analyze the correlation between C.coccoides subgroup,Desulfovibrio,amino acids and insulin resistance.Resluts:1.after 5 weeks for th-e of intervention to decrease random blood glucose,fasting insulin levels,fasting glu-cose level and plasma total cholesterol in insulin resistance mice(P<0.05),Reducing the oral glucose tolerance test and insulin tolerance test blood glucose area under the curve(P<0.05),Reducing insulin resistance index(P<0.05),elevated insulin sensit-ive index(P<0.05);Compared with normal glucose tolerance and insulin resistance group,Gatifloxacin administration group were reduced levels of C.coccoides subgroup,Desulfovibrio in intestinal(P<0.05).Compared with normal glucose tolerance group,There was no significant different between quantity of Plasma amino acids in NGT and Gatifloxacin administration group(P>0.05),the levels of aspartic acid,arginine,thre-onine,serine,alanine,methionine,isoleucine in insulin resistance group were elevated(P<0.05),Glycine concentration decreased(P<0.05).Compared with insulin resistance g-roup,levels of aspartic acid,threonine,isoleucine concentration decreased in Gatifloxa-cin administration group,Glycine concentration increased(P<0.05).2.the level of C.co-ccoides subgroup,Desulfovibrio was positively correlated with the levelsof body we-ight,fasting blood glucose,random blood glucose,fasting plasma insulin,insulin resisa-nce index(r=0.321,P<0.01;r=0.216,P<0.01;r=0.381,P<0.01;r=0.485,P<0.01;r=0.155,P<0.01;r=0.294,P<0.01;r=0.310,P<0.01;r=0.284,P<0.01;r=0.198,P<0.01;r=0.354,P<0.01;r=0.484,P<0.01;r=0.237,P<0.01;r=0.298,P<0.01;r=0.313,P<0.01),and it was negative cor-related with the levels of insulin resistance index(r=-0.305,P<0.01;r=-0.292,P<0.01).the level of C.coccoides subgroup was positively correlated with the levels of aspar-tic acid,serine,threonine,proline,isoleucine(r=0.462,P<0.01;r=0.457,P<0.01;r=0.288,P=0.029;r=0.290,P=0.027;r=0.314,P=0.018),and it was negative correlated with the levels of glycine(r=-0.400,P<0.01);the level of Desulfovibrio was positively correlated wit h the levels of spartic acid,glutamicacid,threonine,alanine,cystine,isoleucine(r=0.529,P<0.01;r=-0.383,P<0.01;r=0.387,P<0.01;r=0.487,P<0.01;r=0.264,P=0.046;r=0.274,P=0.037),and it was negative correlated with the levels of glycine(r=-0.383,P<0.01);Die o-r not,C.coccoides subgroup,glutamic acid has a direct effect on body weight(direct menstruation coefficients were 0.711,0.284),Serine has a direct effect on body weight in mice(directly drift diameter coefficient is 0.225),diet or not the intestines of mice by influencing the C.coccoides subgroup,levels of plasmaserine indirectly influ-ence body weight in mice(indirect drift diameter coefficient is 0.160),C.coccoides subgroup also can by influencing the levels of plasma serine indir-ectly influence b-ody weight in mice mice(indirect drift diameter coefficient is 0.054).Conclusion:Gatifloxacin may be able to affect the levels of C.coccoides subgroup and Desulfovibrio,which affects the plasma amino acids,body glucose and lipidmetabolism;There is association between C.coccoides subgroup,Desulfovibrio,Plasma amino acids and insulin resistance.
