Object:Research Meicha total flavonoids and Meicha protein on blood pressure of SHR and explore its mechanism of action,provide the basis for future clinical application.Method:1.Effect of total flavonoids and Meicha Meicha protein on blood pressure in SHR1.1 The impact of a single dose of blood pressure in SHR rats35 healthy SHR rats were randomly divided into seven groups of five:the model group,Meicha total flavonoids?low,medium and high dose?group,Meicha protein?low and high dose?group,compound Apocynum tablets groups of five.Adaptive feeding rats and blood pressure measurement training1W,the day 9:00 to start the experiment,a group of daily monitoring of blood pressure changes in rats before and after intragastric gavage gavage capacity of 10 mL·kg-1 body weight,model fed rats given volume of distilled water,Meicha total flavonoids low,medium and high dose groups doses were90mg·kg-1,180mg·kg-1,360mg·kg-1,low-protein group Meicha high dose group doses were 70mg·kg-1,140 mg·kg-1,compound Apocynum piece group dose of 0.05g·kg-1,every 15min after administration to measure the blood pressure in rats.1.2 Multiple doses impact on the administration of blood pressure in SHRHealth spontaneously hypertensive?SHR?rats?7-week-old male,SPF grade?70,were randomly divided into 7 groups:model group,Meicha total flavonoids?low,medium and high dose?group,Meicha protein?low and high dose?group,Compound Kendir Leaves Tablets groups,n=10.Adaptive feeding rats and blood pressure measurement training 3W,the model group were orally given volume of distilled water,gavage capacity of 10 mL·kg-1 body weight,Meicha total flavonoids low,medium and high dose groups doses were90mg·kg-1,180mg·kg-1,360mg·kg-1,Meicha protein low andhigh-dose group doses were 70mg·kg-1,140 mg·kg-1,Compound Kendir Leaves Tablets group dose of 0.05g·kg-1,The rats orally administered twice daily gavage seven weeks,blood pressure was measured weekly and record each group.Before the last administration 12h,only water is available.1h after the last administration,draw off the blood from carotidand,keep serum specimens for check;After drawing off the blood,renal and cardiac tissue should be stripped off rapidly.Dry them off by filter paper and weigh precisely,then calculate organ index?organ index=organ weight/body weight?.Take part of the tissue with 4%paraformaldehyde for HE staining.observethe morphology under an optical microscope.2.Research Meicha total flavonoids and Meich protein on the mechanism of the antihypertensive effect of SHRGrouping and method of administration same with 1.2.1 h after the last administration carotid artery blood,sacrificing content of NO,ET-1 and GSH-PX,SOD,MDA serum,blood rapidly after blunt dissection of renal and cardiac tissue,blotted with filter paper water,stored at-80?to take part of the organization,as a real-time PCR test samples to detect renay tissue TLR-4,IL-6,TNF-?and cardiac tissue ACE,Ang?,the expression of CYP11B2.Results:?1?Single dose experiment results showed that:each administration group can lower blood pressure in SHR rats after 30min after administration,where in the antihypertensive effect of the most obvious about 30min,the whole process can be continued down aboout 75min.?2?Multiple doses experiment results showed that:compared with the model group,Meicha total flavonoids?low,medium and high dose?group,Meicha protein?low and high dose?group,compound Apocynum piece group can significantly reduce blood pressure in SHR rats.Although the impact of organ coefficient Meicha Meicha total flavonoids and protein in rats was not significant,but the pathological examination can be found Meicha total flavonoids high dose group,high dose group Meicha protein and compound Apocynum tablets group of SHR rat kidney,heart tissue structure with some protective effect.?3?Serological test results showed that:compared with the model group,Meicha total flavonoids?medium and high dose?group,Meicha protein high-dose group and compound Apocynum tablets,there NO were significantly increased?P<0.01?,and ET-1 were significantly lower?P<0.01?;compared with the model group,Meicha total flavonoids?low,medium and high dose?group,Meicha protein?low and high dose?group and compound Apocynum piece group increased levels of GSH-PX and SOD vitality?both P<0.05?;compared with the model group,Meicha total flavonoids?high dose?group,Meicha protein high-dose group and compound Apocynum piece group,serum MDA content decreased significantly?P<0.01?.?4?Real-time PCR results showed that:compared with model group,Meicha brass total high-dose,medium-dose,high dose of protein TLR-4,IL-6,the expression of TNF-?,ACE,Ang?,CYP11b2 of mRNA was significantly lower?P<0.05?.Conclusion:1.Meicha total flavonoids and protein Meicha single dose can both lower blood pressure in SHR rats,maintained for about 1h antihypertensive effect; Multiple doses experiment can significantly reduce blood pressure in SHR rats,and can improve renal and cardiac tissue pathological changes.2.Meicha total flavonoids and protein Meicha antihypertensive mechanism may be relevant to increase its serum content of NO,reducing the content of ET-1 in serum,reduced mRNA expression of ACE and Ang?cardiac tissue,while its obvious antioxidant and anti-inflammatory effects.
