Quality Evaluation And The Trestment Of Hepatitis Research Of Gentiana Manshurica Fingerprints From Different Origins In The Northeast Regions

Obj ectiveThrough the bars of the Extraction of Gentiana manshurica,quality standards studies,including the study of gentian fingerprints and ultimately determining the advantage origin of the three northeastern provinces;Through the histopathological observation of hepatitis treatment in rats,laiding the foundation for the treatment of hepatitis as Gentiana manshurica clinical research.MethodsUltrasonic extraction method to gentiopicroside content measurement indicators,multi-factor orthogonal experiment to inspect;gentiopicroside as reference,established by HPLC northeast Article Gentiana different habitats Fingerprints chromatographic conditions and determine its content gentiopicroside;to histopathology and body weight changes as an indicator,study the therapeutic effect was gentian extract of hepatitis.ResultsFingerprint:Chromatographic conditions:Column Spherisorb-C18(4.6mm × 250mm,5 ?m),mobile phase:water(A),methanol(B);elutionconditions:0?5min,0?40%B;5?15min,40?60%B;15?25min,60?80%B;25?3 0min,80?100%B;30?40min,100?1 00%B;flo wrate:1.0 mL/min;column temperature:30 ?;injection volume:1 0 ? L;detection time;40min;detection wavelength:270nm.The results show that its total peak relative retention time RSD(%)less than 3%,in line with regulations.In this experiment,as the control gentiopicroside peak,identified a total of 13 peaks,and the total peak relative retention times more stable,but the total peak area greater difference;Optimization of extraction conditions:the main order of the extracted factors were:extraction time(min)>ethanol concentration>extraction times>ethanol consumption,the best optimization conditions:75%ethanol,10 times the amount of ultrasonic extraction one times,the extraction time was 30min;Quality Standards:gentiopicroside Northeast different areas vary,namely:Liaoning Province>Jilin Province>Heilongjiang Province;Pharmacodynamic Study:gentian extract hepatitis has a therapeutic effect.ConclusionThrough the Gentiana manshurica study of the three northeastern provinces(Heilongjiang,Liaoning,Jilin),obtaining the highest content of active ingredients in Liaoning origin,and it is suitable for mass planting gentian;the result of anti-hepatitis experiments shows that gentiana extract hepatitis on rats have a good therapeutic effect,and analysis of gentian provides a basis for treating hepatitis.
