The Shenge formula was composed with Ginseng Radix,Puerariae Radix and Trichosanthis Radix.It was an effective formula for the treatment of type 2 diabetes,combined with Renshen decoction、Yuhu pills and opitimization by pharmacology.Our subject covers preparation process,quality standard,pharmacodynamics mechanism and acute toxicity test of Shenge tablets on DM.Fillers,disintegrants,moistening agent and lubricants were determined by appearance of tablet,hardness and disintegration time.So the best prescription and preparation process were determined.The characteristic compositions were taken as the indexs with TLC to identify the quality of prepared Ginseng Radix and Puerariae Radix.The contents of Puerarin and Ginsenoside Rbl、Ginsenoside Re、Ginsenoside Rgl in the tablets were determined with high performance liquid chromatograpy(HPLC).The results showed that preparation process was security,the quality was controllableThe high fat diet were fed then the streptozotocin in small dose was given to make diabetes models in rats.Glucose metabolism,lipid metabolism,oxidative stress and diabetic nephropathy were investigated in four aspects.The results indicated that Shenge tablets effectively increased body weight and decreased food intake and fluid intake.FBG,oral glucose tolerant test and serum insulin of diabetic rats were ameliorated.After the treatment of Shenge tablets,treatment groups decreased TC,TG,LDL-C levels and the content of MDA in liver tissue of diabetic rats.While HDL-C levels,the content of SOD,GSH and CAT,GSH-PX and GR activity were significantly increased.The high dose group could decrease the higher serum creatinine(Scr)and blood urea nitrogen(BUN)level.The above indicated that Shenge tablets could improve insulin resistance of type Ⅱ diabetic rats by adjusting glucose metabolism and lipid metabolism.Renal impairment and oxidative stress of type Ⅱ diabetic rats were promoted in some degree.The acute toxicity test results showed that the maximum dose of Shenge tablets was given in an average of 20g mice equivalent to 327 times of the clinical adult dosage.So Shenge tablets were safety in clinical use.
