People are increasingly concerned about their own health with the progress of society and economic development.The regulation of human body function with food has become a popular means of the people.Anthocyanins are water-soluble natural pigments,antioxidant function of them has been extensively studied.But the current intake of anthocyanins is still the way to eat.There are few studies on the preparation process,which limits the application.Therefore,the study of anthocyanin preparations has broad market prospects.Black rice contains cyanidin-3-glucoside as the main component of anthocyanins.In this study,black rice anthocyanin was used to develop black rice anthocyanin tablets,and its preparation technology and quality standards were studied.In this paper,the results are as follows: First,the property of materials: High humidity condition and strong light irradiation condition have great impact on the stability of black rice anthocyanin,high temperature condition has very small impact on its stability,The time required for the decomposition of black rice anthocyanins at room temperature for 10% was 210 days.Fe2+,Fe3+ and Cu2+ significantly decreased the stability of black anthocyanin solution,Fe3+ had the greatest effect,Mg2+ had the effect of increasing the stability of black anthocyanin solution.Acidic condition is favorable for the stability of black rice anthocyanin,and the lower the pH value of the solution,the more stable the black rice anthocyanin.Material itself is poor mobility?angle of repose ?=61.07°,loose density ?loose=0.235,tap density ?real=0.407,compression=42.26%?,and the material belongs to "wetting."Second,preparation of the process: The best ratio of black rice anthocyanin and diluent is 1:1,the best ratio of starch and dextrin is 1:1.5,the best amount of disintegrating agent?PVPP?is 2%,the best concentration of ethanol is 75%.Black rice anthocyanin molding preparation process: Black rice anthocyanins and the accessories were passed 100 mesh nylon sieve.All materials are weighed according to prescription.The black rice anthocyanin and diluent and 1/2 of the disintegrant are mixed evenly,the tartaric acid is dispersed in wetting agent.Made soft material,thenpass 20 mesh nylon sieve,then drying the particles under the conditions of 45?,then pass 20 mesh sieve,Fine powder and magnesium stearate and another 1/2disintegrant are mixed evenly,then mixed with particles,then make tablets.Third,quality standards: This product is dark purple convex round tablets,the surface is complete and smooth.The identification method was high performance liquid chromatography,which use cyanydin-3-O-glucoside as the reference substance.3 batches of black rice anthocyanin tablets are all qualified.The content of cyanidin-3-O-glucoside in the preparation was determined by high performance liquid chromatography.The number of theoretical plates is 3947,the reproducibility is good?RSD=1.32%?,the sensitivity is high?The limit of detection is 0.2 ?g/mL,the limit of quantification is 1 ?g/mL.?.There are no delay or smearing of the peak?Tailing factor T=0.97?.The resolution is 2.41,which is over 1.5.The specificity of the method is strong.The precision is high?Intra-day precision RSD=0.37%,Daytime precision RSD=0.63%?.The recovery rate is 78.89%,and the content remains stable within 12 h?RSD=0.76%?.So,the method is suitable for the determination of cyanidin-3-glucoside in the preparation.In addition,the total black rice anthocyanin content in the preparation was determined by the color method.The precision of the method was high?Intra-day precision RSD=0.87%,daytime precision RSD=1.19%?.The recovery rate is 81.71%,and the content remains stable within 12 h?RSD=1.29%?.So,the method is suitable for the determination of anthocyanin in the preparation.Fourth,determination of antioxidant activity in vitro: Black rice anthocyanin and their preparations are showing strong free radical scavenging ability and antioxidant capacity in vitro.The preparation process did not significantly affect the free radical scavenging ability.Finally,the initial stability of the tablets: The effect of temperature on the stability of black rice anthocyanin tablets is small,and the time required to decompose10% at room temperature is 427 days.The stability of the black rice anthocyanin tablets is relatively low under the conditions of high humidity and high light.And in the accelerated test,the stability of 3 batches of black rice anthocyanin tablets decreased with time.In summary,the preparation process of black rice anthocyanin tablets is practicable,and the quality control method is scientific,reasonable and feasible.
