The study was designed to compositae Filifolium sibiricum the whole plant,also known as rabbit tarragon,Siberian tansy,boil poison flower,scared grass,horsetail grass,rabbit hair,boil poisonous for the study,building Filifolium sibiricum fingerprints.Resistant Staphylococcus aureus(MRSA),Escherichia coli,Staphylococcus aureus,Staphylococcus,Salmonella typhi,vice beta were used as pharmacodynamic models to obtain quantitative characteristics of chemical fingerprint and pharmacodynamic data.On this basis,the use of multiple linear regression statistical analysis,to establish a "chromatogram-effect"relationship and speculate Filifolium sibiricum ingredient in antibacterial activity to explore ways to find quickly and efficiently from natural products active ingredient.1 The antibacterial activity of the effective parts of Filifolium sibiricum in vitro experiments.They were taken in June,July,August,the same month fresh,dried,flowers,no flowers were extracted and separated to give 14 samples,and twenty-six monomer component,using broth dilution method and drilling method to Resistant Staphylococcus aureus(MRSA),Escherichia coli,Staphylococcus aureus,Staphylococcus,Salmonella typhi,vice beta model five kinds of bacteria,the samples were measured for each sample antibacterial activity,having their views on the five bacterial inhibition zone diameter and minimum inhibitory concentration(MIC).The results showed that 14 batches of sample of Filifolium sibiricum antibacterial effects were different,Filifolium sibiricum inhibition of bacterial five descending order are:Resistant Staphylococcus aureus,Staphylococcus,Staphylococcus aureus,Escherichia coli,deputy B Salmonella typhi.Antibacterial activity is:August>July>June,no flowers>flowers,dried>fresh.2 HPLC chemical fingerprint of Filifolium sibiricum was Established.HPLC conditions were optimized,according to the requirements of chemical fingerprint,chromatographic fingerprint evaluation system software of Chinese medicine promulgated by state pharmacopoeia commission was used to establish and evaluat HPLC fingerprint of Filifolium sibiricum antibacterial activity part.The results showed that Filifolium sibiricum on HPLC fingerprint similarity established are up 90%,can be seen in each sample total peak area of difference,suggesting that medicinal ingredients are different,and therefore also the difference inhibitory effect.3"Chromatogramm-activity"relationship model of Filifolium sibiricum was constructed.Statistical analysis using multiple linear regression method,the Filifolium sibiricum HPLC fingerprint of a total peak area to quantify the relative peak areas obtained with the drilling method,of 14 batches of Filifolium sibiricum antibacterial activity inhibition zone diameter results in combination,antimicrobial herbs leaf daisy building line "spectrum"-"efficacy" relationship,suggesting that Filifolium sibiricum related to its activity in the effective part of a larger chemical composition.Using multiple linear regression showed that:Filifolium sibiricum HPLC fingerprint of 18 fingerprint peaks,(1)Resistant Staphylococcus aureus role had nine peaks,were No.2,3,4,6,8,9,10,12,18 peak,the regression coefficient R2 = 0.928 seen their total contribution rate of 92.8%;(2)Escherichia coli had nine peaks,were No.2,3,4,5,10,11,14,16,18 peaks,the regression coefficient R2 = 0.914 seen their total contribution rate of 91.4%;(3)Staphylococcus aureus had nine peaks,were No.2,3,4,5,10,11,14,16,18 peaks,the regression coefficient R2 = 0.852 seen their total contribution rate of 85.2%;(4)Staphylococcus has nine peaks,were No.2,3,4,5,10,11,14,16,18 peaks,the regression coefficient R2 = 0.902 seen their total contribution rate of 90.2%;(5)The sub B Salmonella typhi role has nine peaks,were No.2,3,4,5,10,11,14,16,18 peak,the regression coefficient R2= 0.922 seen their total contribution rate of 92.2%.Verification result 4 Filifolium sibiricum samples indicates the resulting "Spectrum-effect"relationship between calculated and measured actual value is very close.4 The concentration of Filifolium sibiricum effective monomer component parts was 1 mg· mL-1,(1)Resistant Staphylococcus aureus exhibit strong antibacterial activity are:isorhamnetin-3-O-?-D-glucoside,mango glycosides,scopolamine glycosides,scopoletin,quercetin,kaempferol,caffeic acid.(2)Exhibit antibacterial strong activity against E.coli.(3)Against Staphylococcus aureus exhibit strong antibacterial activity are:Myricetin,isoquercitrin,isoorientin,rutin,mango glycosides,kaempferol,hyperin,hydroxytyrosol.(4)Staphylococcus exhibit strong antibacterial activity are:Myricetin isorhamnetin-3-O-?-D-glucoside,rutin,mangiferin,ferulic acid,scopoletin,kaempferol,Isorhamnetin.(5)The sub B Salmonella typhi exhibit strong antibacterial activity are:scopolamine glycosides,luteolin strong antibacterial activity.(6)hydroxyacetophenone phthalate,di-n-butyl ester,among trimethyl pyrogallol,east atropine,phloroglucinol did not exhibit antibacterial activity.
