Objective:Screening out effective Chinese herbal compound W for anti-small melon worm disease,preparation of the best extracts of traditional Chinese herbal compound W,for using in prevention and control of Small melon worm.Methods:1 Establishing pharmacodynamic evalustion method,W animals infected with the parasite Small melon as experimental animals,to screen the 13 traditional Chinese medicine prescription in insecticidal test.2 Establishing the ultraviolet spectrophotometry chemical evaluation method,to study extraction method,comminution degree,dosage of solvent,extraction time and extraction times of the compound W,optimizing the various single factor level,using the L₉?3⁴?orthogonal experiment with the optimum extraction technology of compound W.3 Extract 1 g/ml of the best extract of traditional Chinese herbal compound W.4 According to the safety test of test animals with different concentrations of compound C extracts,the average mortality rate of the test animals was converted into probability units,Bliss method was used to calculate and analyze the LC50 of the animals at 24,48,and 96 h and statistics by the probit probability unit analysis method in the spss20.0software.5 The butanol-acetic acid-water?4:2:1?as developing agent,bismuth potassium iodide solution as the chromogenic agent,using thin layer chromatography?TLC?to authenticate quality of the best extract of traditional Chinese medicine compound W.6 The mobile phase:acetonitrile-water?28:72?;detection wavelength:205 nm,using the HPLC detect of content of index ingredients H in the best extract of compound W.7 Taking pharmacodynamics test to evaluate 1 g/ml of the best extract of traditional Chinese herbal compound W.Results:The crude extract of traditional Chinese herbal compound W in 60/ml,72 h to kill Small melon is 100%,and Small melon did not die;The ultraviolet absorption maximum wavelegth of compounfW extract is475 nm,the RSD value of linearity,repeatability,stability,sample recovery is less than 2%;The optimum extraction process of compound W:crushing through 60 mesh sievem selection of solvent 75%ethanol,reflux extraction 2 times,each time 60 min,dosage of extraction solvent for 10 times every time;The TLC conditions of butanol-acetic acid-water?4:2:1?is feasible and the spots of the reference substance solution,reference substance medicinal material sample solution and instructions of test sample solution are clearly visible,the Rf value is 0.55;In pharmacodynamics experiment,the concentration of the best extract of traditional Chinese herbal compound W is 40mg/L,the mortality of Small melon is 100%in 53 h,during the observation of 96 h,experimental animals without exception.Conclusion:The best extract of comound W is safe and effective to Small melon worm,can provide the basis for the development of new drugs.
