Acne is a chronic inflammatory skin disease that often occurs in the pilosebaceous glands during adolescence and is mainly characterized by papules,comedones,cysts,nodules,pustules,and scars.Because of its strong beauty and high incidence,it has become a disease of increasing public concern,so it is necessary to give certain drug intervention treatment.Modern medical treatment is not ideal,and side effects,so there is an urgent need for safer and more effective treatment;and traditional Chinese medicine?TCM?has significant advantages in the treatment of acne.The external treatment of TCM has its unique advantages in acne treatment,not only to avoid the irritation of oral drugs to the gastric mucosa,but also convenient to use.There are many kinds of external TCM for treating acne,and the therapeutic effect of compound prescription composed of single herbs with synergistic or additive effect is better than that of single herbs.Abnormal microbial proliferation plays an important role in the development of acne,and the clinical pathogenesis of acne in TCM is mainly manifested by the heat accumulation of the lung meridian,the interaction of toxic heat and the accumulation of damp heat.Therefore,it is necessary to conduct a reasonable prescription and compatibility study on the traditional Chinese medicine with good antibacterial or bactericidal effect on acne-related pathogenic bacteria.Galla Chinensis has anti-bacterial,anti-inflammatory,anti-oxidant,anti-viral,anti-aging and other pharmacological effects,among which gallotannic acid's astringent,bacteriostatic and anti-inflammatory effects are closely related to the treatment of acne,so gallotannic acid and its compound treatment Acne has certain research value.In this paper,through the exploration of the etiology and pathogenesis of acne and the excavation of TCM literature for acne treatment.According to the characteristics of acne syndrome differentiation and treatment,with S.epidermidis and E.coli as the object,screen out the effective compound which has the functions of astringing and congestion,detoxification and carbuncle,promoting blood circulation and removing blood stasis..The main research methods and results are as follows:1.Comparative study on screening methods of TCM and determination of minimum inhibitory concentration?MIC?.Through the preliminary screening of the relevant literatures on the treatment of acne,eight kinds of traditional Chinese medicines,including Scutellaria baicalensis,Coptis chinensis,Red peony root,Taraxacum officinale,Morus alba,Morus alba L,Sophora flavescens and Cortex Moutan were obtained in addition to gallotannic acid,and S.epidermidis and E.coli were used as test strains for antibacterial experiments.The results showed that the MIC determined by in vitro double dilution method was more sensitive than the filter paper method.The antibacterial effect of gallotannic acid on S.epidermidis and E.coli was stronger than Tara tannic acid,and the comprehensive antibacterial ability of the nine Chinese herbal extracts except Tara tannic acid against the two kinds of bacteria was gallotannic acid>Scutellaria baicalensis>Taraxacum officinale>Moutan bark>Chishao>Coptis chinensis>Morus alba L>Morus alba>Sophora flavescens.2.Research on the optimal compatibility of gallnut compound.According to the compatibility principle of TCM formula and various Chinese herbal extracts comprehensive prescription antibacterial ability of pathogenic bacteria associated with acne,and taking gallotannic acid,Scutellaria baicalensis,Taraxacum officinale,Cortex Moutan,Radix Paeoniae Rubra and Rhizoma Coptidis as test raw materials.The filter paper method was used for in vitro antibacterial test of acne-related pathogenic bacteria,and the inhibition zone diameter was used as an evaluation index for orthogonal test optimization to determine the optimal compatibility ratio of gallnut compound.The results showed that Galla Chinensis compound with the concentration of Radix Paeoniae Rubra,Coptis chinensis,gallotannic acid,Radix Scutellariae,Taraxacum officinale,and Cortex Moutan extract in the ratio of 3.6:1.25:1.0:5.0:2.5:0.75 had the best antibacterial effect on E.coli and S.epidermidis,and its inhibition zone diameters were 9.69 mm and 19.34 mm,respectively.The compound takes Galla Chinensis as the monarch drug,Taraxacum officinale and Coptis chinensis as the minister drug,Scutellaria baicalensis as the adjuvant drug,Cortex Moutan and Radix Paeoniae Rubra as the enabling drug,and the whole prescription has the effects of clearing away heat and toxic substances,promoting blood circulation to remove blood stasis,and astringent sore muscle.3.Study on the bacteriostatic effect of Gallnut nectar combined with optimal gallnut compound.The antibacterial activity of Gallnut nectar and Bauhinia honey was determined by Oxford cup method,and the antibacterial activity and MIC of the new compound after Gallnut nectar was combined with the optimal gallnut compound were determined by filter paper method and tube double dilution method,respectively.The results showed that the inhibitory activity of Gallnut nectar against both bacteria was stronger than Bauhinia nectar,and 20%Gallnut nectar could enhance the antibacterial effect of the optimal gallnut compound,The inhibition zone diameters of the compound of Gallnut nectar dosage form against E.coli and S.epidermidis were 10.11 mm and21.68 mm,respectively,and the MICs were 22.03 and 1.38 mg/m L,respectively.The results showed that Gallnut nectar,as an excipient of the compound,not played a role in harmonizing various drugs,but enhanced the antibacterial activity of the optimal gallnut compound.4.The antibacterial mechanism of Gallnut nectar dosage form compound on the main pathogenic bacteria of acne was preliminarily explored.The effects of Gallnut nectar dosage form compound on bacterial extracellular K+,Mg2+,AKP and intracellular total protein content were determined by spectrophotometer.The results showed that the contents of extracellular K+,Mg2+and AKP were increased,and the synthesis of total intracellular protein was decreased.Therefore,it is speculated that the compound of Gallnut nectar dosage form compound mainly increases its permeability and permeability by destroying the structure of cell wall and cell membrane of E.coli and S.epidermidis,resulting in the leakage of intracellular substances and the decrease of protein synthesis,and then inhibits the growth of bacteria.In this study,for the first time,Galla Chinensis and Gallnut nectar,which have the characteristics of Wuling Mountain area,were scientifically screened and compatible under the guidance of the compatibility theory of the traditional Chinese medicine formula"Jun-Cheng-Zuo-Zhuo",and finally the best compatibility formula with good antibacterial effect on acne-related pathogenic bacteria was obtained.Through mechanism study,it was found that the compound mainly inhibited the growth of bacteria from many aspects by destroying the structure of cell membrane and cell wall of main pathogenic bacteria of acne,increasing its permeability and permeability leading to the leakage of intracellular substances,and at the same time,the synthesis of intracellular proteins decreased,indicating that the antibacterial mechanism of the compound was more comprehensive.It lays a certain foundation for the future development of external preparations for clinical treatment of acne,the development of functional skin care products and the depth mechanism research of the compound.
